姜黄素对挤压综合征大鼠肾损伤及JNK信号通路活化抑制作用的实验研究 被引量:1

Experimental Study on Inhibition Effect of Curcumin on Renal Injury and Activation of JNK Signaling Pathway in Rats with Crush Syndrome

摘要 目的:观察姜黄素对挤压综合征大鼠肾损伤和JNK信号通路活化的抑制作用。方法:重物挤压建立大鼠挤压综合征模型,实验分为正常组、模型组、姜黄素低剂量组、姜黄素高剂量组,造模前2 h腹腔注射给药,观察各组大鼠肾组织及肌肉组织的病理变化,在造模6、12、24 h分别检测肾功指标(BUN、SCr),Western blot法检测各组大鼠肾组织JNK信号通路活化水平。结果:除正常组外,其余各组大鼠均可见肌肉和肾脏损伤,姜黄素组损伤较模型组为轻。与正常组比较,各造模组血清BUN、SCr水平明显升高(P<0.05),肾组织p-JNK表达明显增强(P<0.05),随挤压伤时间延长呈加重趋势;姜黄素组血清BUN、SCr水平较模型组降低(P<0.05),肾组织p-JNK表达明显减弱(P<0.05),高剂量组效果更显著。结论:姜黄素可以有效地减轻挤压综合征大鼠肾损伤,抑制肾组织JNK信号通路活化。 Objective :To investigate the effect of Curcumin on renal injury and activation of JNK signaling pathway in rats of crush syndrome. Methods : Models with crush syndrome were built by heavy extrusion, experi- ment was divided into normal, model and Curcumin low, high dose groups, Curcumin was given to rats by intrap- eritoneal injection before modeling 2 hours. The pathological changes of crushed muscle and kidney were ob- served, serum BUN and SCr levels were determined after modeling 6,12,24 hours, c - jun protein expression in kidney tissue was determined by Western blot method. Results:The muscle and kidney injuries were obvious in modeling rats except normal group, those in Curcumin groups were slighter than those in model group. To com- pare with normal group, the serum BNU and SCr levels were significantly raised in modeling rats, p - JNK ex- pression in renal tissue was enhanced significantly, these changes got worse with time ; serum BNU and SCr levels of Curcumin groups were lower than those in model group, p - JNK expression in renal tissue was abated, the effects of high dose group were more significant ( P 〈 0.05 ). Conclusion: Curcumin can effectively reduce kidney injury in Crush syndrome rats,inhibit the activation of JNK signal pathway in renal tissue.
