摘要 以驼峰藤(Merrillanthus hainanensis)带芽茎段为外植体,研究不同消毒方法及植物生长调节剂对其离体培养及植株再生的影响,建立了驼峰藤离体快繁技术体系。结果表明,带芽茎段的最佳消毒灭菌方法为75%乙醇浸泡20 s,0.1%HgCl_2浸泡10 min;腋芽诱导的最适宜培养基为MS+2.00 mg/L 6-BA+0.10mg/L NAA+0.10 mg/L GA_3,诱导率可达81.11%;适宜腋芽增殖的培养基为MS+3.50 mg/L 6-BA+0.15mg/L NAA,增殖系数可达6.24;最佳生根培养基为1/2MS+0.30 mg/L NAA,生根率为84.44%,该结果为驼峰藤的快繁提供了一条新途径。 In order to establish the tissue culture and rapid propagation system of Merrillanthus hainanensis,the effects of different disinfection method and plant growth regulators on plant regeneration in vitro were studied using stems with axillary buds as explants. The results showed that the better disinfection method for stems with axillary buds was to use 75% alcohol immersion 20 s,then use 0.1% HgCl_2 immersion 10 min;the fittest medium for germination of stem axillary bud was MS+2.00mg/L 6-BA +0.10 mg/L NAA +0.10 mg/L GA_3,with a 81.11% germination rate of axillary bud. The best medium for shoot multiplication was MS +3.50 mg/L 6-BA +0.15 mg/L NAA,the multiplication coefficient could reach 6.24. The best rooting medium was 1/2MS+0.30 mg/L NAA,with a 84.44% rooting rate. The research supplied a new way for rapid propagation of Merrillanthus hainanensis.
