摘要
【目的】阿勒泰羊是新疆北部地区重要的产多羔绵羊品种,但一直未找到影响其高繁殖力的主效基因。以影响排卵数的BMP15基因作为候选基因,从分子水平上对阿勒泰的多胎机制进行研究。【方法】以PCR直接测序法获得SNP位点,分析突变位点的特性,并通过大规模的群体检测统计推断其遗传效应。【结果】阿勒泰羊在BMP15基因的内含子667 bp和外显子2的755 bp处发生突变,2个位点均只有2种基因型,且纯合子为优势基因。与产羔数关联分析发现仅外显子2的755 bp处位点2种基因型差异显著,经群体验证得到同样结果,TC型母羊比TT型母羊平均多产0.8羔。同时该处的突变引起了氨基酸由亮氨酸向脯氨酸变化。【结论】故推测BMP15基因与控制阿勒泰羊多胎性能的主效基因存在紧密的遗传连锁。
[ Objective] Aletai sheep was an important breed of sheep in the northern of Xinjiang China, but the main genes affecting its prolif-icacy had not been found. In order to study the multiple mechanism, the BMP15 gene which affected ovulation was taken as a candidategene, the fecundity mechanism at the molecular level of Aletai was researched. [ Method ] SNP loci were obtained by direct sequencing ofPCR, the characteristics of its mutations were analyzed, and its genetic effect through the mass detection statistics was inferred. [ Result ] The667 bp of the intron and the 755 bp of the exon 2 in BMP15 gene in Aletai sheep had mutation, and the two loci had only two genotypes.The analysis associated with litter size found that only the two genotypes of 755 bp of exon 2 had significant difference, which were obtainedby the same group experience card, and TC ewes than TI ewes average prolific 0.8 lambs. At the same time, its mutation caused the changeof amino acid from leucine to proline. [ Conclusion]Therefore, the BMP15 gene was closely related to the control of the main gene of Metalsheep.
