摘要
【目的】探明地鳖肽对四氯化碳(CCl_4)诱导的小鼠急性肝损伤的保护作用。【方法】取60只小鼠分溶剂组、模型组(CCl_4)、地鳖肽组、阳性药物组,灌服不同剂量地鳖肽提取液和生理盐水及保肝药,每天1次连续7d,末次给药2h后除溶剂组小鼠外都腹腔注射0.1%CCl_4,制备血清和肝组织匀浆,ELISA和PCR等方法检测血清中ALT和AST活力,肝组织中SOD、GSH-Px、CAT活力和MDA含量,组织中TNF-α、IL-6和iNOS含量及mRNA表达;显微镜观察肝组织结构。【结果】与模型组比较,地鳖肽组小鼠血清中ALT和AST活力极明显降低,肝组织中抗氧化酶SOD、CAT、GSH-Px活力显著提高,但其MDA含量明显降低;肝组织中TNF-α、IL-6和iNOS含量极显著降低及mRNA表达量明显下调;模型组小鼠肝组织结构损伤明显,而地鳖肽组小鼠肝组织结构损伤明显缓解。【结论】地鳖肽对CCl_4诱导的小鼠急性肝损伤有保护作用。
[Objective]The experiment was to investigate the protective effect of ESW peptides against CCl4-induced acute liver injury in mice. [Methods]60 mice were randomly divided into vehicle group, model group(CCl4 ), ESW peptides group, posi- tive drugcontrol group, and they were given relevant medicine intragastrically once a day, and continuously for 7 days. All mouse except for those in vehicle group were induced by intraperitoneal injection of 0.1% CCl4 to acute liver injury 2hr after the last administeration. Serum and liver from the mice were prepared. Activity of ALT, AST, SOD, GSH-Px, CAT and content of MDA were determined, and the content and mRNA expression of TNF-α, IL-6, iNOS were evaluated by ELISA and RT- PCR respectively. HE staining was used to detect the histopathology of the liver. [Results]Compared with the model group, the activity of ALT and AST in ESW peptides group were significantly decreased, the activity of antioxidant enzymes SOD, CAT and GSH-Px were significantly increased but the content of MDA liver tissue was decreased in ESW peptides group. There was a significant decrease in the content and mRNA expression of TNF-α, IL-6, iNOS in ESW peptides group compared with model group. According to the observation of liver tissue histopathology, the obvious damage in liver were observed in model group, ESW peptides could obviously alleviate degree of liver injury. [Conclusion]ESW peptides showed a protective effect on the acute liver injury of CCl4-induced mouse.
