摘要
目的 24 h尿蛋白测定是评判肾功能的“金标准”,而尿液标本的合理储存是获取准确结果的前提.曱苯和二曱苯 均可作为防腐剂用于24 h 尿蛋白的测定,但关于二者防腐效应的比较研究还鲜见报道,本文试图从生物安全和经济性方面分析 两者的优劣.方法 收集并立即检测未洛加任何防腐剖的24 h 尿液标本,选择即刻测定尿蛋白浓度约为1500 mg/L 的标本针对 防腐剂、储存时间、储存温度因素按照正交试验设计分组,分别测定不同储存条件下标本尿蛋白浓度.通过极差分析、方差分析获 得优化储存条件.结果 0. 5%二曱苯防腐效果最好,其次为0. 5%曱苯和0. 1%二曱苯,且三者均优于未添加防腐剂对照组.最 佳储存温度和时间分别为25 ° C和4 h.综合考虑生物安全性、经济性和实用性,将防腐条件优化为在常温(25°C)条件下使用0. 1%二曱苯储存24 h 尿液标本检测尿蛋白,此条件在检测限范围内均取得较好的线性和偏倚.结 论 较 低浓度(0.1 % )的二曱苯 可以作为24 h 尿蛋白检测的防腐剂,尤其适合住院患者标本的储存.
Objective 24 hour urinary protein detection is a gold standard for estimating the renal function, but the precondition of obtaining the accurate results depends on the proper samples storage. Both toluene and xylene can preserve the sample of 24 hour urinary protein, however the comparative report between them is still rare. This study is to evaluate the their advantage and disad-vantage in the aspect of biological safety and economical efficiency. Methods After Collecting and detecting the urinary samples for 24 hours without addition, We classified the 24-hour urinary protein sample which the concentration was 1500 mg/L into groups following to orthogonal experimental design in the aspect of preservative, storage time and storage temperature, then the samples in each group was analyzed, finally the storage condition was optimized by refering to the method of variance analysis. Results 0. 5% xylene had a good preservative effect prior to 0. 5 % toluene or 0. 1 % xylene, moreover this three were better than the control group without preservative. Optimal storage temperature and time were respectively 25°C and 4h, however because of the biologi-cal safety, economy and practicability, we optimized the storage condition which the 24-hour urinal protein samples were stored at room temperature (25 ° C) with 0. 1% xylene as preservative. And the optimized condition showed the excellent linearity and bias 〇- ver the detection limit. Conclusion The lower concentration (0. 1%) xylene was competent for the storage of 24-hour urinal pro-tein as a preservative, and was particularly suitable for in patient specimens.
