摘要
目的:探讨黄芪多糖对ABCA1蛋白表达的影响。方法:将RAW264.7巨噬细胞诱导分化成泡沫细胞,用不同浓度黄芪多糖对其干预24h,酶法测定泡沫细胞内胆固醇酯含量,ELISA法检测细胞内ABCA1的蛋白表达。结果:对照组、10、20、50、100mg/L的APS作用于泡沫细胞24h后,细胞内胆固醇酯含量分别为45.15±3.57、37.67±2.26、34.59±3.51、28.57±10.34、15.92±3.86nmol/mg。各组ABCA1蛋白浓度为(6.02±6.12)、(15.86±9.15)、(54.76±14.20)、(75.39±10.09)、(97.56±13.25)ng/L。各APS组与对照组比较,差异具有统计学意义(P<0.01)。结论:黄芪多糖可能通过上调ABCA1的蛋白表达而降低泡沫细胞内胆固醇含量。
Objective:To investigate the effect of APS on the protein expression of ABCA1 in RAW264.7 macrophage-derived foam cells.Methods:RAW264.7 macrophages were incubated with APS(0,10,20,50,100mg/L).The cholesterol content in foam cells was determined by enzyme digestion with different concentrations of astragalus polysaccharides for 24 h.The expression of ABCA1 was detected by ELISA.Results:In the control group,the contents of cholesterolester in foam cells were(45.15±3.57),(37.67±2.26),(34.59±3.51),(28.57±10.34),(15.92±3.86)nmol/mg respectively after the application of APS(0,10,20,50,100mg/L)for 24h(P0.01).The concentration of protein in each group was(6.02±6.12),(15.86±9.15),(54.76±14.20),(75.39±10.09),(97.56±13.25)ng/L.Compared with the control group,the difference was significantly significant.Conclusion:APS may reduce the cholesterol content of foam cells by up-regulating ABCA1 protein expression.
