摘要
目的:探讨葛根素对人肾小管上皮细胞(HK-2)缺氧复氧(HR)损伤的保护作用及机制。方法:以人肾小管上皮细胞株HK-2为研究对象,建立细胞HR模型,分为对照组、HR组、HR+葛根素组。倒置显微镜观察细胞形态,四甲基偶氮唑蓝(MTT)法检测细胞活力,丙二醛(MDA)试剂盒检测细胞MDA含量,乳酸脱氢酶(LDH)试剂盒检测细胞LDH释放量,超氧化物歧化酶(SOD)试剂盒检测细胞SOD活性,Western-blotting法检测细胞bax、bcl-2蛋白的表达水平。结果:葛根素可减少HR性损伤HK-2的MDA生成及LDH释放,增强SOD活性,上调bcl-2蛋白的表达,下调bax蛋白的表达。结论:葛根素对HK-2的HR性损伤有明确的保护作用,其机制与降低细胞氧化应激损伤和减少细胞凋亡有关。
Objective: To investigate the protective effect of puerarin against hypoxia-reoxygenation (HR) injury in human renal tubular epithelial HK-2 cells and possible mechanisms. Methods: Human renal tubular epithelial HK-2 cells were used to establish a cell model of HR and were divided into control group, HR group, and HR+puerarin group. Inverted microscopy was used to observe cell morphology, methyl thiazolyl tetrazolium assay was used to evaluate cell viability, malondialdehyde (MDA) kit was used to measure MDA level, lactate dehydrogenase (LDH) kit was used to measure the amount of LDH released by cells, superoxide dismutase (SOD) kit was used to measure SOD activity, and Western blot was used to measure the protein expression of Bax and bcl-2 in cells. Results: Puerarin reduced the production of MDA and the release of LDH in HK-2 cells with HR injury, increased the activity of SOD, upregulated the expression of bel-2, and downregulated the expression of Bax. Conclusions: Puerarin exerts a marked protective effect against HR injury in HK-2 cells, possibly by re- ducing oxidative stress injury and inhibiting ceil apoptosis
