NE在COPD患者肺组织中的表达及其与巨噬细胞凋亡的相关性研究

Expression of neutrophil elastase in lung tissue of patients with chronic obstructive pulmonary disease and its association with macrophage apoptosis

目的:观察中性粒细胞弹性蛋白酶(neutrophil elastase,NE)在慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)患者肺组织中的表达及其与巨噬细胞凋亡的相关性。方法:收集因肺部肿瘤行肺叶切除手术患者肺组织60例,其中非COPD患者(A组)30例,COPD者(B组)30例。采用免疫组织化学方法检测NE在肺组织中的表达;采用TUNEL法检测肺组织中巨噬细胞凋亡率,采用髓过氧化物酶染色法(MPO)检测巨噬细胞的染色阳性率。结果:B组肺组织中NE的蛋白表达较A组明显增强(P<0.05);B组肺组织中巨噬细胞凋亡率较A组明显降低(P<0.05);B组肺组织中巨噬细胞MPO染色阳性率较A组明显降低(P<0.05);B组NE的蛋白表达量与肺功能指标(FEVl%,FEVl/FVC%)呈负相关,与巨噬细胞凋亡率及MPO染色阳性率呈负相关。结论:NE在COPD患者肺组织中的表达上调并与巨噬细胞凋亡的相关,NE可能参与了COPD的气道炎症过程,也是引起气流阻塞的重要因素。

Objective：To investigate the expression of neutrophil elastase （NE） in the lung tissue of chronic obstructive pulmonary disease （COPD） patients and its association with macrophage apoptosis. Methods：Lung tissue samples were collected from 60 patients who underwent pulmonary lobectomy due to lung tumor, and among these patients, 30 had no COPD （group A） and 30 had COPD （group B）. Immunohistochemistry was used to measure the expression of NE in lung tissue, TUNEL was used to measure maerophage apoptosis rate, and myeloperoxidase （MPO） staining was used to measure the positive rate of MPO staining in maerophages. Results： Compared with group A, group B had a significant increase in the expression of NE in lung tissue and significant reductions in macrophage apoptosis rate and positive rate of MPO staining in macrophages （P 〈 0.05）. In group B, the protein expression of NE was negatively correlated with pulmonary function parameters FEVI% and FEV1/ FVC%, macrophage apoptosis rate, and positive rate of MPO staining in macrophages. Conclusions：The expression of NE is upregulated in lung tissue of COPD patients and is associated with maerophage apoptosis. NE may be involved in airway inflammation and is also an important factor for airway obstruction.
