正交试验优化葶苈生脉方中醇提工艺研究

摘要 优选葶苈生脉方中红参、五味子、川芎、葶苈子、泽泻的最佳提取工艺,通过正交试验设计考察乙醇浓度、料液比、提取时间对工艺的影响,采用高效液相色谱法(HPLC)进行梯度洗脱,以人参皂苷Rg1和人参皂苷Rb1为指标进行综合评分,确定最佳提取工艺:6倍量65%乙醇,回流提取3次,第一次3 h,第二次2 h,第三次1 h。通过验证实验表明该工艺科学、合理、稳定、可行,为葶苈生脉方开发为现代中药制剂提供科学依据。 The best extraction technology of Ginseng, Schisandra, Chuanxiong, Tingling and Alisma oriental in Tingli Shengmai decoction was optimized. Three factors of ethanol concentration, ethanol dosage and extraction time were investigated by the orthogonal test. The separation was performed by HPLC. The method of gradient elution extraction was used to determine the content of ginsenoside Rgl and ginsenoside Rbl in solution, the sum of the two contents was used as a comprehensive evaluation index to confirm the optimum extraction process. The best extraction method was as follows: 6 times of 65% ethanol, 3 times of reflux, 3 h for the first time, 2 h for the second time and 3 h for the third time. The extraction process is scientific, reasonable, stable and feasible, and it can provide the scientific basis for the development of Tingli Shengmai decoction of becoming the modern Chinese medicine.
