摘要
目的:研究SMCC在不同溶液中的降解情况。方法:将SMCC分别用ACN、DMA和DMSO等有机溶剂溶解并制成一定浓度(约3 mg/ml)的储备液,再用不同的缓冲液稀释10倍并混匀作为供试品溶液,采用高效液相色谱法观察主峰的降解情况。结果:对于SMCC的水解,碱性环境以及DMSO或DMA的存在有显著的促进作用,而酸性环境以及ACN的存在则有一定的抑制作用。结论:交联反应可在弱碱环境下以DMA/DMSO作溶剂,而色谱分析则可在酸性环境下以ACN作溶剂。
Objective: To investigate the degradation phenomenon of SMCC in different solutions. Methods: Stock solutions of SMCC were prepared in ACN, DMA and DMSO, respectively, to give a final concentration of 3 mg/ml. Working solutions were prepared by 10-fold dilution of stock solutions in several buffer solutions. The degradation of SMCC was monitored by an HPLC method. Results: The hydrolysis of SMCC could be accelerated with the introduction of basic buffer solution or DMA/DMSO, while it could be hindered with the introduction of acidic solution or ACN. Conclusion: Cross-linking reaction could be performed in weakly alkaline condition by using solvent of DMA/DMSO, while HPLC analysis could be carried out in acidic condition by using solvent of ACN.
