水溶性红雪茶多糖体外结合胆酸盐能力的分析 被引量:5

摘要 [目的]红雪茶多糖在小鼠体内的降脂保肝作用已有学者证实,本研究主要通过不同处理方法提取红雪茶多糖,分析其体外结合胆酸盐能力,从而探讨其降血脂机理。[方法]利用水提醇沉法,结合去蛋白和不去蛋白处理,用6种不同浓度乙醇溶液提取得到12种红雪茶多糖提取液,通过分光光度法计算反应后多糖溶液中甘氨酸胆酸钠和牛磺酸胆酸钠的含量,分析多糖对胆酸盐的结合能力。[结果]结果表明:1.不同处理条件下的红雪茶多糖对胆酸盐均有结合能力,且随着多糖浓度的增加,其对胆酸盐的结合能力逐步增强;2.去蛋白后的红雪茶多糖对胆酸盐的结合能力比不去蛋白的强;3.红雪茶多糖对牛磺酸胆酸钠的结合能力要强于多糖对甘氨酸胆酸钠的结合能力。[结论]红雪茶多糖具有体外结合胆酸盐的能力,可能是其具有降脂保肝功能的重要机理之一。 [Objective] Lethariella spp. polysaccharide had been approved to have lipid-lowering and hepatoprotective effect in mice by some scholars. In this study, the bile salt-binding ability of Lethariella spp. polysaccharide in vitro was analyzed to explore its lipid-lowering mechanism. [Methods] Twelve kinds of polysaccharide extracts were obtained by water extraction and alcohol precipitation method, combining with or without protein-removed processing, using six different concentration of ethanol. The content of glycine sodium cholate and taurine sodium cholate in the polysaccharide solution was calculated respectively by spectrophotometry. [Results] 1. Lethariella spp. polysaccharide had the binding ability of bile salt, and the binding ability improved gradually with the increase of polysaccharide extracts' concentration. 2, The bile salt-binding ability of protein-removed polysaccharide extracts was stronger than that of protein non-removed polysaccharide extracts. 3. The binding ability of polysaccharide to bind taurine sodium cholate was stronger than that of polysaccharide to bind glycine sodium cholate. [Conclusion] Lethariella spp. polysaccharide had the binding ability of bile salt in vitro, which may be one of the key mechanism related to lipid-lowering and hepatoprotective function.
