摘要
目的观察水蛭素对喉癌荷瘤小鼠的抑瘤作用。方法将40只小鼠随机分为4组,每组10只,小鼠左前腋窝下皮下种瘤建立实体瘤模型,阳性对照组以氟尿嘧啶20mg/kg腹腔注射;水蛭素高剂量组以2.5g/(kg·d)灌胃;阴性对照组以生理盐水0.1m L/(10g·d)灌胃;水蛭素低剂量组以1.2g/(kg·d)灌胃。连续用药14 d后处死小鼠取荷瘤鼠肿块称重,并将瘤组织包埋、固定、切片、染色等,计算抑瘤率,利用Image-Pro Plus 6.0图像分析软件检测瘤组织中C-met的表达。结果水蛭素高、低剂量组对小鼠喉癌细胞均有明显的抑制作用,抑瘤率分别为40.1%、26.92%,与对照组比较差异有统计学意义(P<0.05);4组中C-met均呈阳性表达,水蛭素高、低剂量组C-met的表达均下调,与阴性对照组比较差异有统计学意义(P<0.05)。结论水蛭素对小鼠喉癌细胞抑制作用显著。
Objective To investigate the inhibitory effect of hirudin on the laryngeal carcinoma tumor - bearing mouse. Methods 40mice were randomly divided into 4 groups (n = 10) ,and planted with laryngeal cancer cells subcutaneous at the left armpit to set the tumor model. In positive control group, mice were intraperitoneal injected with fluorouracil (20 mg/kg) ;in negative control group,mice received NS 0. 1ml/ ( 10 g · d) ,in high - dose bimdin group,mice were accepted 2.5 g/(kg · d). While low - dose hirudin group received 1.2 g/( kg · d). Mice were sacrificed after continuously treatment for 14 days, and the tumor were taken and weighed, the tumor tissue were prepared for immunostaining to calculate the tumor inhibition rate and the expression of C - met. Results Between the high - dose and low - dose of hirudin inhibited the laryngeal cancer cells, the tumor inhibition rate was 40.1%, 26.92% respectively, which had significant difference compared with control group ( P 〈 0.05 ), C - met positive staining was observed in 4 groups while the expression was down- regulated in both high- dose and low- dose hirudin groups,which had significant difference compared with control (P 〈 0.05). Conclusion The inhibitory effect of hirudin is significant on the laryngeal carcinoma tumor - bearing mice.
