摘要
对白痢沙门菌二元调控系统响应蛋白CpxR进行生物信息学分析,提示CpxR可能发挥了重要的生理功能。同时,构建了重组表达载体pET32a-CpxR,转化表达菌株BL21后,对其进行诱导表达,SDS-PAGE和Western blot结果表明CpxR获得了正确表达。纯化蛋白免疫小鼠制备了多抗血清,Western blot验证其有一定的免疫原性。为深入研究其生理功能和调控机制奠定了理论基础。
Pullorum disease(PD) is an acute systemic disease with high morbidity and mortality in young chicks, and can be transmitted to chicks through eggs. Consequently, PD is difficult to eradicate and seriously threats poultry industry. Salmonella Pullorum (S. Pullorum), the causal agent of PD,is a classical facultative intracellular bacteria and encounters many kinds of complicated hos- tile surroundings in vivo. In this research, the bioinformatics analysis of CpxR hints that CpxR maybe play an important physiological function. Meanwhile, the recombinant expression vector pET32a-CpxR was contructed and transformed into the expression strain BL21 for inducible ex- pression. SDS-PAGE and Western Blot analysis showed that a confirmed CpxR was obtained. After immunized mice,the serum against CpxR was isolated,and proved that CpxR had a good immunogenicity by Western blot. This research provides a foundation for further study on the physiological function and regulatory mechanism of CpxR from S. Pullorum.
