石油污染土壤中漆酶活性的影响因素 被引量:4

Influence factors of laccase activity in oil-contaminated soil

摘要 针对纯化酶生产成本较高且土壤中漆酶活性较低的问题,通过20 d的室内对照实验,采用ABTS分光光度法测定漆酶活性,研究Cu^(2+)、pH、温度、碳氮比及含水率对漆酶活性的影响,结果表明5个环境因素的最优值分别是40 mg·kg^(-1)、5.2、30℃、40∶1和20%。环境因素与漆酶活性的高低有关,其相关性从大到小依次为pH、温度、含水率、碳氮比和铜离子浓度。其中,pH的相关性最高,相关系数为0.956。在最优条件下,培养30 d后石油烃降解率从自然条件下的8.54%提高到18.79%,最高漆酶活性达到13.97 U·g^(-1)。 Based on the problem that the production cost of purified enzyme is higher and the laccase activity in soil is lower,using ABTS spectrophotography to determine laccase activity,to study the effects of copper concentration,temperature,pH,carbon-nitrogen rate and percentage of moisture on laccase activity,through a 20-days laboratory experiment.The results showed that,the optimal values of five environmental factors were 40 mg ·kg^-1,5.2,30 ℃,40 ∶ 1 and 20%.There had relationship between environmental factors and laccase activity,the correlations from high to low were pH,temperature,water content,carbon-nitrogen ratio and copper concentration.Among them,the correlation of pH was the highest and its correlation was 0.956.After 30 d the hydrocarbon degradation rate was raised to 18.79% compared with degradation rate under natural condition which was8.54%,and the highest laccase activity was 13.97 U·g^-1.
