摘要 目的观察小鼠肾脏发育过程中泌尿小管中整合素连接激酶(ILK)的表达变化。方法培育各日龄小鼠,采用免疫组织化学结合体视学方法及Western blotting技术检测胚龄(E)12 d、14 d、16 d、18 d胎鼠及生后日龄(P)1 d、3 d、7 d、14 d仔鼠肾组织中ILK的表达及含量变化。结果 ILK在E12 d即开始表达于输尿管芽细胞,随后表达在各期肾小管和集合管的上皮细胞胞质中。体视学测量和Western blotting检测结果显示ILK的表达随着肾脏的发育逐渐增强,在P7 d达到高峰,随后无明显变化。结论 ILK在小鼠肾泌尿小管的发育过程中存在时空性表达规律。 Objective To observe the expression of integrin -linked kinase (ILK) in urinary tubules during the development of mouse kidney. Methods Mice were nurtured and divided into embryo (E) 12 d, 14 d, 16 d, 18 d and postnatal (P) 1 d, 3 d, 7 d, 14 d. The expression and content of ILK were examined by immunohistochemistry, stereologieal methods and Western blotting. Results ILK began to express in ureteral bud cellsat El2 d, then it expressed in cytoplasm of renal tubular and col- lecting duet epithelial cells. Results of stereology and Western blotting analysis showed that the level of ILK increased gradually with the development of the kidney. It reached the peak at postnatal day 7, and then remained stable. Conclusion The expressions of ILK in the development of renal urinary tubules of mice demonstrated temporal and spatial sequence. ILK might affect the differentiation and development of renal urinary tubules epithelial cells.
