Simultaneous determination of six constituents in Qishiwei Zhenzhu Pills by HPLC

目的建立HPLC法同时测定藏药七十味珍珠丸(西红花、降香、甘草等)中6种成分的含有量。方法该药物50%甲醇提取液的分析采用Inertsil■ODS-3色谱柱(250 mm×4.6 mm,5μm);以0.2%磷酸-乙腈为流动相,梯度洗脱;体积流量0.8 m L/min;柱温30℃;检测波长254 nm。结果没食子酸、柯里拉京、沉香四醇、鞣花酸、西红花苷Ⅰ、西红花苷Ⅱ分别在1.41~42.24、0.61~18.24、0.30~9.12、0.47~14.04、0.62~18.48、0.32~9.45μg/m L范围内线性关系良好(R2≥0.999 4),平均加样回收率(RSD)分别为93.41%(1.75%)、96.84%(1.75%)、97.45%(0.58%)、93.22%(0.56%)、97.01%(1.39%)、97.22%(1.11%)。2个厂家不同批次样品中各成分含有量有一定差异,尤其以柯里拉京和沉香四醇更明显。结论七十味珍珠丸质量不稳定,应引起关注。

AIM To establish an HPLC method for the simultaneous content determination of six constituents in Tibetan medicine Qishiwei Zhenzhu Pills （Croci Stigma,Dalbergiae odoriferae Lignum,Glycyrrhizae Radix et Rhizoma,etc.）.METHODS The analysis of 50% methanol extract of this drug was carried out on a 30 ℃ thermostatic InertsilODS-3 column （250 mm × 4.6 mm,5 μm）,with the mobile phase comprising of 0.2% phosphoric acid-acetonitrile flowing at 0.8 m L/min in a gradient elution manner,and the detection wavelength was set at 254 nm.RESULTS Gallic acid,corilagin,agarotetrol,ellagic acid,crocinⅠ and crocin Ⅱ showed good linear relationships within the ranges of 1.41-42.24,0.61-18.24,0.30-9.12,0.47-14.04,0.62-18.48 and 0.32-9.45 μg/m L （R^2≥0.999 4）,whose average recoveries （RSDs） were 93.41% （1.75%）,96.84% （1.75%）,97.45% （0.58%）,93.22% （0.56%）,97.01% （1.39%） and 97.22% （1.11%）,respectively.The contents of various constituents in different batches of samples from two manufactures showed some differences,especially for those of corilagin and agarotetrol.CONCLUSION We should pay attention to the unstable quality of Qishiwei Zhenzhu Pills.
