雷帕霉素下调基质金属蛋白酶表达抑制伯基特淋巴瘤细胞转移的作用和机制研究

摘要 目的:研究雷帕霉素(Rapamycin,RAPA)对伯基特淋巴瘤细胞株Raji中基质金属蛋白酶(MMP-2,MMP-9)表达水平的影响,探讨雷帕霉素在伯基特淋巴瘤细胞株Raji转移方面的作用。方法:以伯基特淋巴瘤细胞株Raji为实验对象,应用Transwell迁移实验、Western blot、qPCR技术来检测雷帕霉素对Raji细胞迁移能力,MMP-2、MMP-9的蛋白和mRNA表达水平的抑制情况。结果:雷帕霉素可显著抑制Raji的迁移能力,且呈浓度依赖性,当雷帕霉素浓度为1 nmol/L时抑制作用最强(P<0.05);Raji细胞中MMP-2、MMP-9蛋白及mRNA的表达水平有明显的下降趋势,且呈浓度依赖性,mRNA的表达水平在作用48 h时最低。结论:雷帕霉素可以显著抑制伯基特淋巴瘤细胞株Raji的迁移能力,降低MMP-2、MMP-9的蛋白及mRNA的表达,抑制淋巴瘤细胞的转移。 Objective:To study the effect of Rapamycin (RAPA) on the expression of MMP-2 and MMP-9 in Burkitt lymphoma cell line Raji,and to discuss the influence of RAPA on the migration of Burkitt lymphoma cell line Raji in vitro.Methods:Using Transwell model to examine the migration ability of Raji.Using Western blot and qPCR to examine the expression of protein and mRNA of MMP-2 and MMP-9 in Raji cells.Results:Rapamycin significantly inhibited the migration ability of Raji in a concentration-dependent style and the largest inhibitory effect of rapamycin was at 1 nmol/L (P〈0.05).Rapamycin inhibited the expression of protein and mRNA of MMP-2 and MMP-9 in a concentration-dependent style,and the minimum expression of mRNA of MMP-2 and MMP-9 occurred at 48 hours.Conclusion:Rapamycin significantly inhibited the migration ability of Raji,and reduced the expression of protein and mRNA of MMP-2 and MMP-9 in Raji cells,and inhibited the migration of Burkitt lymphoma cell line Raji.
