山羊附睾头细胞转染中新霉素和嘌呤霉素的浓度筛选 被引量:2

摘要 通过对9月龄黑山羊附睾头细胞进行不同浓度、不同时间新霉素和嘌呤霉素的处理,收集各个时间的细胞并进行计数,旨在研究山羊附睾头细胞对新霉素和嘌呤霉素的耐受浓度。结果显示,山羊附睾头细胞在转染时对新霉素和嘌呤霉素适宜的质量浓度分别为400,2μg/m L;试验细胞计数结果与细胞生长状态相一致,山羊附睾细胞与其他哺乳动物不同组织器官对新霉素和嘌呤霉素的耐受浓度基本一致。研究结果可为后期基因敲除和过表达载体转染进山羊附睾头细胞时抗性基因浓度的确定提供了参考,也为后期基因功能的研究奠定了基础。 This research was designed to study the tolerance of goat epididymal cells to neomycin and puromycin. The cells of different time were collected and counted at different concentrations of neomycin and puromycin. The results showed that the appropriate concentrations of goat epididymal cells to neomycin and puromycin were 400, 2 μg/m L, respectively. The results of this test cell count consistent with the cell growth status, and the tolerance to neomycin and puromycin of goat epididymal cells and other mammalian cells of different tissues and organs was consistent. The results of this study will provide a reference for gene knockout and the determination of resistance genes of overexpression vector transfection into the goat epididymal cells, and to provide a basis for the study of the gene function.
