海参ACE抑制肽活性HPLC检测方法的建立

摘要 建立测定海参体壁ACE抑制肽活性的HPLC分析方法.采用反相高效液相色谱法,色谱柱为Zorbax SB-C18分析柱(4.6×250 mm,填料粒径5μm);流动相:乙腈/超纯水(体积比为1:3);流速:1 m L/min;检测波长:228 nm,柱温:25℃;进样量:10μL.结果显示马尿酸浓度在0~800μg/m L范围内,浓度与其峰面积呈现良好的线性关系(R2=0.9994),最低检测限为0.2μg/m L.该法具有较好重复性(RSD 2.12%)和稳定性(RSD1.31%),经海参体壁酶解液和依那普利检测验证,该方法可用于检测海参体壁ACE抑制肽活性. A high performance liquid chromatographic method to determine angiotensin I-converting enzyme inhibitor activity by sea cucumber was established. ZorbaxSB-C18 column (4.6×250 mm, 5μm) was treat- ed with mobile phase acetonitrile-water (1:3). The content was detected at 228 nm. The flow rate was 1.0 mL/min. The results showed that the linear range was 0-800 μg/mL, the linear correlation coefficient was 0.9994, and the detection limit was 0.2 μg/mL. The method has good reproducibility (RSD2.12%) and stabil- ity (RSD1.31%). This method should be suitable for detection of ACE activity from sea cucumber through enzymatic hydrolysis of sea cucumber and enalapril.
