重度低龄儿童龋进展中唾液多肽的初步研究

A preliminary study of salivary peptidome profiling analysis for the progression of severe early childhood caries

摘要 目的检测重度低龄儿童龋(severe early childhood caries,s-ECC)进展过程中唾液多肽表达水平的变化。方法纳入3~5岁s-ECC儿童、无龋儿童各6名。分别于基线、基线后一个月及基线后四个月收集刺激性全唾液,采用弱阳离子交换磁珠联合基质辅助激光解吸/电离飞行时间质谱技术检测样本中多肽表达水平差异,BioExplorer v1.0软件分析数据。结果筛出随龋病进展表达差异有统计学意义的3个多肽峰(P<0.05),m/z分别为4612、2894.2、4064.2Da,多肽峰4612和2894.2随龋病进展表达水平呈上升趋势,4064.2呈下降趋势。结论重度低龄儿童龋进展中,唾液中存在表达差异的多肽峰,为日后深入研究提供一定的理论依据。 Objective The aim of the present study is to detect the discriminating salivary peptidome profiles during caries progression using stimulated whole saliva of children at the age of 3 ~5 years with severe early childhood caries(s-ECC). Methods According to the inclusion criteria, 12 children aged 3~5 years were recruited to attend the present study. 6 of them were clinically diagnosed as s-ECC and grouped as the experimental group, and the other 6 were caries-free and grouped as control. Stimulated whole saliva sample of each subject was collected individually at each time point(baseline, after 1 month, after 4 month). Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS)was used to detect the expression levels of peptides, whilst Bio Explorer software was used to analyze the data. Results In the longitudinal observation of the s-ECC samples, three peptides peaks(m/z values: 4612, 2894.2,4064.2Da) showed significant difference during the development of s-ECC. Conclusion Salivary peptides of children with s-ECC showed significant difference during caries progression, which provide a theoretical basis for further research.
