红景天苷对大鼠心房钠尿肽分泌的影响

Effects of salidroside on the secretion of atrial natriuretic peptide in rats

[目的]探讨红景天苷对大鼠心房钠尿肽(ANP)分泌的影响.[方法]采用大鼠离体搏动的心房灌流模型,利用蛋白激酶C(PKC)的β,δ及ε(PKCβ,PKCδ,PKCε)阻断剂观察红景天苷对大鼠心房ANP分泌的影响.采用放射免疫法测定ANP含量.[结果]与对照组比较,红景天苷明显增加心房ANP的分泌(P<0.01);PKCβ亚型阻断剂LY333531和PKCε亚型阻断剂EAVSLKPT均可完全阻断红景天苷增加ANP分泌的效应,而PKCδ亚型阻断剂Rottlerin则部分减缓红景天苷对ANP分泌的效应.[结论]红景天苷明显增加心房ANP的分泌,作用机制认为是通过PKC信号通路的激活实现的.

OBJECTIVE To investigate the effects of salidroside on the secretion of atrial natriuretic peptide in rats.METHODS The isolated beating atrial perfusion model of rats was adopted,and the blockers of protein kinase C（PKC）β,δandε（PKCβ,PKCδ,PKCε）were used to observe the effects of salidroside on the secretion of rat atrial natriuretic peptide.Radioimmunoassay was used to measure the content of ANP.RESULTS Compared with the control group,salidroside obviously increased the secretion of ANP（P〈0.01）.LY333531 and EAVSLKPT,blockers of PKCβand of PKCε,could completely block the effects of salidroside on increasing ANP secretion,while rottlerin,blocker of PKCδ,partly slowed down the effects of salidroside on the secretion of ANP.CONCLUSIONSalidroside significantly increased the secretion of ANP,and the mechanism might be through the activation of PKC signaling pathway.
