姜黄素对癫痫幼鼠学习记忆及神经元凋亡的影响 被引量:2

摘要 目的观察姜黄素对癫痫幼鼠的学习记忆及海马组织神经元凋亡的影响并探讨其机制。方法将50只21 d龄的SD幼鼠随机分正常组,模型组,姜黄素高(100 mg/kg)、姜黄素中(50 mg/kg)、姜黄素低(25 mg/kg)剂量组,姜黄素各组给予相应剂量姜黄素灌胃,连续2周。Morris水迷宫法检测乳鼠学习记忆能力、TUNEL法检测乳鼠海马神经元的凋亡、Western blotting法检测Cleaved-caspase-3蛋白的表达情况。结果与模型组大鼠比较,姜黄素高、中、低剂量组的学习记忆能力显著提高(P<0.01);细胞凋亡情况显著改善(P<0.01);且Cleaved-caspase-3蛋白的表达显著降低(P<0.01)。结论姜黄素可能通过抑制Cleaved-caspase-3蛋白的表达,从而抑制癫痫幼鼠海马神经元的凋亡。 Objective: To explore the effects of curcumin on learning and memory and neuron apoptosis in epileptic immature rats. Methods: A total of 50 male SD rats around 21 days of age were randomly allocated into5 groups: the normal saline group,pentylenetetrazole(PTZ) group,high-dose curcumin group(100 mg/kg),middle-dose curcumin group(50 mg/kg),and low-dose curcumin group(25 mg/kg). Curcumin was administrated orally for 2w. Morris water maze method was used to detect the learning and memory ability of neonatal rats;TUNEL method was used to detect the apoptosis of hippocampal neurons and the expression of cleaved-caspase-3protein was detected by Western blotting method. Results: Compared with in the model group,the high,medium and low dose groups significantly improve the ability of learning and memory(P 〈 0.01);cell apoptosis was significantly improved(P 〈 0.01);the expression of cleaved-caspase-3 protein was significantly decreased(P 〈 0.01).Conclusion: Curcumin can inhibit the expression of cleaved-caspase-3 protein and inhibit the apoptosis of hippocampal neurons in epileptic rats.
