胶质和粉质重楼总皂苷的超声提取工艺及活性成分的含量差异研究 被引量:3

摘要 目的通过超声提取工艺及HPLC法比较胶质和粉质重楼总皂苷及其皂苷单体的含量差异。方法以总皂苷得率为指标,正交试验优化重楼总皂苷的超声提取工艺后,按照优选的工艺对6批次重楼总皂苷、皂苷单体进行HPLC含量测定。色谱条件:Agilent C18柱(4.6mm×250mm,5μm),流动相:乙腈-水,梯度洗脱:0~40min,30%~65%乙腈,流速:0.8ml·min-1,检测波长:203nm。结果在超声提取条件下,最佳工艺为:乙醇浓度60%,料液比1∶50,提取时间为30min,胶质重楼总皂苷含量高于粉质重楼;6种皂苷单体HPLC检测条件下标准曲线线性良好(r>0.9998),粉质重楼6种皂苷单体含量与胶质重楼存在显著差异。结论建立的方法准确、可靠、稳定,为重楼药材的质量控制提供科学依据,并为胶质和粉质重楼的合理使用提供了数据支持。 Objective To compare the content difference of total saponins by ultrasonic extraction and by high performance liquid chromatography between colloidal Paris polyDhylla and amyloid Paris polylhylla .Methods The content of total saponins was chosen as the experimental index, and orthogonal design was used to optimize the conditions of ultrasonic extraction of total saponins.The column was Agi- lent C18(250 mm×4.6 mm,5 μm),the mobile phase was acetonitrile and water with gradient elution (0- 40 min 30%-65% acetonitrile) at a flow rate of 0.8 ml .min-l.The detection wavelength was 203 nm.Re- suits The optimum ultrasonic extraction conditions were as follows: ethanol concentration was 60 %, solid- liquid ratio was 1 : 50 and extraction time was 30 min.Six steroidal saponins had a good linear relationship (r 〉 0.9998). The results showed that the content of total saponins of colloidal Paris polyphylla was higher than that of amyloid Paris polyphylla ,and that the content of six steroidal saponins was different between different batches. Conclusion This established method is accurate, reliable and stable, which can provide data for quality control and rational use of Paris polyphylla .
