摘要
目的建立多穗柯叶有效部位的指纹图谱。方法针对影响指纹图谱的色谱条件进行考察,确定最佳色谱条件:Kromasil C_(18)柱(250 mm×4.6 mm,5μm);甲醇-0.1%(体积分数)甲酸系统梯度洗脱;柱温为25℃;流速为0.8 m L/min;检测波长为254 nm;进样体积为25μL;采集时间为140 min。测定了10批多穗柯叶药材,以根皮苷为参照峰,确定共有峰,建立多穗柯叶有效部位的HPLC指纹图谱;采用"中药色谱指纹图谱相似度评价系统(2004A版)"进行指纹图谱分析。结果建立了多穗柯叶指纹图谱,共找出了10个共有峰,各样品与自动生成对照图谱的相似度均在0.95以上。结论本方法具有良好的精密度、稳定性和重复性,可作为多穗柯综合质量控制的评价方法。
Objective To develop an HPLC method for the fingerprint analysis for the active part of Lithocarpus polystachyus ( Wall.) Rehd. Methods The chromatographic factors of fingerprints were investigated to optimize the HPLC conditions: C18 reverse phase column Kromasil ( 250 mm × 4. 6 mm, 5 μm) , methanol-0. 1% formic acid gradient elution system, column temperature of 25 ℃, the flow rate of 0. 8 mL/min, the detection wavelength of 254 nm, the injection volume of 25μL, and the acquisition time of 140 min. The common peaks of the 10 batches were selected to establish the HPLC fingerprint of the active part of L. polystachyus with phlorizin as reference. The fingerprint analysis was carried out with the"Chinese medicine chromatographic fingerprint similarity evaluation system ( 2004A )". Results The fingerprint of the L. polystachyus leaves were established, and 10 common peaks were identified with a good separation. The similarity between various samples and the automatically generated reference fingerprints reached above 0.95. Conclusion The method shows good precision, stability and repeatability and could be used for its quality evaluation.
