热休克蛋白(HSP)90是一类ATP依赖性分子伴侣。HSP90可活化并稳定多种客户蛋白,其中许多客户蛋白都涉及重要的信号通路。HSP90影响许多生理过程,如信号转导、胞内运输和蛋白降解,其已成为肿瘤治疗的新靶点之一。HSP90在多发性骨髓瘤(MM)中过度表达,而抑制HSP90可诱导MM细胞凋亡,并抑制其重要的存活和增殖通路。HSP90在MM耐药机制中发挥着重要作用,这使得HSP90抑制剂成为新型的MM治疗药物。
Heat shock protein (HSP)90 is a class of ATP dependent molecular chaperones. HSP90 can activate and stabilize a variety of client proteins, many of which involved in important cellular pathways. HSP90 affects many physiological processes, such as signal transduction, intracellular trafficking and protein degradation, and has become a new target for tumor therapy. HSP90 was found over expression in multiple myeloma (MM). Inhibition of HsP90 can induce MM cell apoptosis and inhibit the proliferation and survival of the important pathway, and HSP90 play an important role in the mechanism of resistance to MM, which makes HSP90 inhibitor become the new drug treatment of MM.
