脾虚1号方对脾虚型功能性消化不良大鼠肝脏细胞色素C氧化酶IV的影响 被引量:5

摘要 目的:探讨脾虚型功能性消化不良(FD)大鼠肝组织细胞色素C氧化酶IV(COX IV)蛋白与基因表达及脾虚1号方干预。方法:70只10 d龄雄性SD大鼠按随机数字表法分为正常对照组(10只)、FD模型组(单模型组10只)、脾虚型FD模型组(50只)。正常组给予2%蔗糖溶液灌胃,FD模型组和脾虚型FD模型组均给予0.1%蔗糖碘乙酰胺蔗糖溶液灌胃,0.2 m L/只·d,连续6 d。脾虚型FD模型组正常饲料喂养至6周龄后叠加改良小平台站立,连续14 d;造模结束后按随机数字表法分为双模型组、多潘立酮组、脾虚1号方低、中、高剂量组各10只,药物干预14 d,采用免疫组化、Western-blot和RT-PCR方法检测肝脏组织COX IV蛋白与mRNA表达量。结果:与正常组比较,双模型组大鼠肝脏COX IV mRNA表达量和平均光密度值显著降低;与双模型组比较,脾虚1号方高剂量组大鼠肝脏COX IV mRNA和蛋白表达量升高。结论:脾虚型FD大鼠存在COX IV mRNA和蛋白表达量降低现象,脾虚1号方可能是通过上调COX IV蛋白与mRNA的表达提高脾虚证大鼠能量代谢。 Objective: To investigate the expression of COX IV protein and mRNA in Liver tissue of Dyspepsia rats with spleen deficiency type and the intervention by Pixu I Recipe. Method: 70 10-day-old male SD rat pups were randomly divided into normal group( n = 10),FD model group( single model,n = 10),FD with spleen deficiency model group( n =50). The normal control group were gavaged with 0. 2 m L of 2% sucrose,meanwhile,the FD with spleen deficiency model group were gavaged with 0. 2 m L of 0. 1% IA in 2% sucrose,for 6 days. To postnatal 43 day,the FD with spleen deficiency model group was given modified multiple platform method( MMPM),continued 14 h every day,for 14 days. The FD with spleen deficiency model group were averagely randomly divided into double model group,domperidone group,low-dose,medium-dose and high-dose Chinese herbs group,and treated by drug for 14 d. To detect the COX IV protein expression in liver tissues by immunohistochemistry,Western-blot and RT-PCR. Results: Compared with the normal group,the mRNA and protein expression of COX IV of double model group were significantly decreased; Compared with the double model group,the mRNA and protein expression of COX IV of high-dose Pixu I Recipe group were increased. Conclusion: The FD Rats with Spleen Deficiency had the phenomenon of low expression of COX IV mRNA and protein,and Pixu I Recipe could increase the expression of COX IV protein and mRNA,and then to promote metabolic energy.
