刺老苞根皮含药血清对原代成骨细胞增殖、分化和矿化功能的影响 被引量:3

Effect of Aralia Echinocauiis Hand. Mazz Containing Serum on Proliferation,Differentiation,Mineralization of Primary Osteoblasts

摘要 目的:研究不同浓度刺老苞根皮含药血清对原代成骨细胞的增殖、分化及矿化功能的影响。方法:SPF级健康雌性SD大鼠(80只)给予蒸馏水、刺老苞根皮水煎剂高、中、低3种剂量浓度,连续灌胃7 d制备含药血清。各组含药血清干预培养成骨细胞,采用微量酶标法检测碱性磷酸酶活性、茜素红染色钙化结节并计数,流式PI单染法检测成骨细胞周期。结果:在干预培养后与模型对照组比较,各剂量组碱性磷酸酶活性、钙化结节数有所升高,中、高剂量组高于模型对照组;与模型对照组比较,流式细胞仪检测发现各剂量组G_0/G_1期、G_2/M期明显下降,S期和G_2/M+S期明显升高。结论:刺老苞根皮含药血清可通过延长原代成骨细胞S期,促进其增殖、分化及矿化功能的作用。 Objectives: Observing the influence of Aralia echinocau Iis Hand. Mazz containing serum on the proliferation,differentiation,mineralization of primary osteoblasts. Methods: SD rats( n = 80) were used to make Aralia echinocau Iis Hand. Mazz containing serum by gastric perfusion for seven days with distilled water,Aralia echinocau Iis Hand. Mazz water decoction high,middle,low dosages. In vitro,the primary osteoblasts were cultured with the drug serum in each group. Detection of alkaline phosphatase activity by trace enzyme method,cells were detected by alizarin red staining experiments,osteoblasts cycle were analyzed by flow cytometry. Results: After osteoblasts were intervened with the serum in each group,compared with model groups,osteoblasts were cultured with drug serum in each group,the alkaline phosphatase activity and the ability of mineralization were improved. The G0/G1 and G2/M phase of the osteoblasts cycle were decreased. The S and G2/M + S phase were increased. Conclusion: Effects of Aralia echinocaulis Hand. Mazz containing serum could extend the S phase of osteoblasts. The proliferation,differentiation,mineralization of primary osteoblasts were promoted.
