摘要
目的利用微针的促渗作用对小鼠进行经皮免疫,探讨不同免疫部位所引起免疫应答的特点。方法用染色法和组织切片法考察微针促渗仪的穿刺能力和穿刺效果;采用微针促渗仪,对小鼠的后背皮肤和耳部皮肤进行处理,涂抹模型抗原卵清蛋白(OVA)和佐剂Cp G ODN 1826后,采用ELISA法测定血清中的抗OVA特异性抗体。结果微针促渗仪可成功穿刺皮肤角质层,将抗原OVA和佐剂Cp G ODN 1826同时递送至皮肤表皮层。与经后背皮肤免疫比较,经耳部免疫会产生更强效的Ig G、Ig G 1和Ig G 2a特异性抗体水平。结论经皮免疫在耳部皮肤引起了比后背皮肤更强的免疫效果。
OBJECTIVE To study the immune responses of different delivery locations by means of microneedle. METHODS The penetration ability of microneedle was evaluated by staining and section was used to characterized transdermal delivery effects. After pretreatment with sold microneedles on the skin of the dorsal side and the ears,the model antigen OVA and adjuvant Cp G ODN 1826 were mixed and dripped onto the treated areas. The serum was collected to analyze the level of anti-OVA specific antibody by ELISA.RESULTS The microneedles we used could successfully penetrate the stratum corneum of the skin and open upmicropores to let antigen OVA and adjuvant Cp G ODN 1826 through the skin at the same time. CONCLUSION Compared with immunization on the dorsal skin,immunization on the ears can induce more potent immune responses and produce more antibody,such as Ig G,Ig G 1 and Ig G 2a.
