刺五加内生放线菌Streptomyces tauricus CWJ-107活性次级代谢产物研究 被引量:4

摘要 目的从药用植物刺五加内生放线菌Streptomyces tauricus CWJ-107次级代谢产物中发现抗耐甲氧西林金黄色葡萄球菌(MRSA)活性良好的天然产物。方法采用16S r RNA基因序列分析对菌株CWJ-107进行初步分类鉴定,大量发酵获得粗提物,通过硅胶柱层析、LH20凝胶柱层析、薄层色谱以及高效液相色谱等方法,以抗MRSA活性导向进行目标化合物的分离纯化,利用核磁共振和质谱鉴定化合物结构,并利用微孔板法测定化合物最小抑菌浓度(minimal inhibitory concentration,MIC),对其抗MRSA活性进行评价。结果菌种鉴定结果显示菌株CWJ-107为Streptomyces tauricus,从菌株CWJ-107的菌丝体中分离得到1个抗MRSA活性化合物107-A,经波谱解析证实其结构为糖基化蒽醌类化合物cinerubin B,抗菌活性测定结果证实其具有良好的抗MRSA活性,MIC值为1μg/m L。结论从刺五加内生放线菌CWJ-107的代谢产物中分离到的107-A经结构鉴定为糖基化蒽醌类化合物cinerubin B,除此之外,本文首次报道了该化合物的抗MRSA活性。 Objective To isolate active natural products with good anti-MRSA activity from the strain Streptomyces tauricus CWJ-107, which was isolated from the medical plant Acanthopanax senticosus. Methods Phylogenetic analyses ofthe strain CWJ-107 were based on 16S rRNA gene sequence. Extractions were obtained from large scale fermentation. Then the target compound was isolated and purified with silica gel column chromatography, LH20 column chromatography, TLC and HPLC, which was guided by anti-MRSA activity. And the structure of the obtained compound was elucidated by NMR and mass spectrometry. Furthermore, the activity of the purified compound against MRSA was assayed by the micro plate method. Results The strain CWJ-107 was identified as Streptomyces tauricus. And an active compound 107-A was isolated from the fermented mycelia of the strain CWJ-107, the structure was determined as glycosylated anthraquinones, cinerubin B. Interestingly, bioassay results showed that compound 107-A exhibited good activity against MRSA (MIC=I μg/mL). Conclusion Compound 107-A, isolated from the fermented mycelia of the Acanthopanax senticosus strain CWJ-107, was identified as glycosylated anthraquinones, cinerubin B. In addition, this paper firstly reports the anti-MRSA activity of this compound.
