摘要 目的采用基因组重排技术选育泰乐菌素高产菌株。方法通过经典诱变育种中获得的增产突变株中选取4株作为基因组重排的出发亲本,对它们进行了3轮基因组重排育种,筛选得到了高产重排菌株,其中1株重排菌株TLA16-336的泰乐菌素产量15.62g/L,比产量最高的亲本菌株TLA15-521(12.53g/L)提高了24.7%,比原始出发菌株TL12-142(11.75g/L)提高了32.9%。结果高产重排菌株在5L发酵罐上进行发酵试验,其泰乐菌素产量为16.03g/L,比原始出发菌株产量13.30g/L提高了20.5%。结论对于泰乐菌素发酵效价的提高,基因组重排是一个可行的方法。 Objective To improve the production of tylosin by genome shuffling. Methods Genome shuffling was applied to improve tylosin-producing by utilizing the four highest strains derived on scripture mutation as the starting population. A recombinant TLA16-336 with improved phenotype was obtained by three rounds of genome shuffling, and its production reached 15.62g/L, which was increased by 24.7% and 32.9% compared with that of the highest parent strain and the original strain, respectively. Results Fermentation experiments by TLA16-336 were conducted in 5L fermentor. The tylosin production of the highest producer mutant reached 16.03g/L, which was increased by 20.5% compared with that of the original strain. Conclusion Genome rearrangement was feasible method for improving fermentation yield.
