芦丁对大鼠心肌缺血再灌注损伤的保护作用 被引量:11

摘要 目的:探讨芦丁对大鼠心肌缺血再灌注损伤的保护作用及机制。方法:将雄性SD大鼠随机分假手术组,缺血再灌注损伤模型组,芦丁高(80 mg/kg)、中(40 mg/kg)、低剂量组(20 mg/kg),共5组,每组10只。结扎冠状动脉左前降支建立心肌缺血再灌注损伤模型,心肌缺血再灌注后超声心动图评估心功能,测定各组大鼠心肌梗死面积,酶标仪检测血清及组织心肌酶生成量和氧化指标活性的变化,HE染色法观察组织细胞形态,免疫印迹法检测血红素加氧酶-1(HO-1)的表达。结果:与模型组比较,芦丁治疗组大鼠左心室射血分数及左心室短轴缩短分数升高,心肌梗死面积减小,血清肌酸激酶同功酶、乳酸脱氢酶、谷草转氨酶水平和丙二醛含量降低,超氧化物歧化酶活性和谷胱甘肽含量升高,心肌内HO-1表达增多(P均<0.05)。结论:芦丁可提高HO-1的表达、减少和清除氧自由基,起到保护心肌缺血再灌注损伤的作用。 Aim: To investigate the cardioprotective effect of rutin on myocardial ischemia-reperfusion injury in rats.Methods: Male SD rats were randomly allocated into Sham operation group,myocardial ischemia-reperfusion group( MI/R group),rutin high dose group( RH group,80 mg/kg),rutin middle dose group( RM group,40 mg/kg),and rutin low dosegroup( RL group,20 mg/kg)( n = 10). Left anterior descending coronary artery was ligated to establish the myocardial ischemia-reperfusion injury model. Echocardiography evaluation of cardiac function after myocardial ischemia-reperfusion injury was performed. The area of myocardial infarction was determined. The myocardial enzymes( CK-MB,LDH,and AST)production and oxidation indexes( SOD,GSH and MDA) activity in serum and tissue were determined with enzyme-labeled instrument. The cell morphology was observed by HE staining. The expression of heme oxygenase-1 enzyme( HO-1) was detected by Western blot. Results: Compared with the MI/R group,EF and FS in the rutin groups were significantly increased,the area of myocardial infaction and myocardial enzymes were significantly decreased,the production MDA was significantly decreased,the activity of SOD and the production of GSH significantly increased,and the expression of HO-1in the rutin groups was markedly increased( P〈0. 05). Conclusion: Rutin could increase HO-1 expression,therefore promote the endogenous antioxidant system against the myocardial ischemia-reperfusion injury in rats.
