摘要
目的通过动物实验,研究异功散早期干预对支气管哮喘幼年小鼠气道炎症的影响,以探索中医药防治支气管哮喘的机制。方法选取7日龄BALB/c品系雌性小鼠24只,随机分为空白对照组、模型对照组和异功散组共3组。以卵蛋白诱导法建立幼年小鼠的哮喘模型,观察幼鼠的一般情况、肺组织病理(HE染色)及肺泡灌洗液(bronchial alveolar lavage fluid,BALF)中白细胞介素10(interleukin 10,IL-10)、转化生长因子-β1(transforming growth factor-beta1,TGF-β1)和IL-17的变化(ELISA法)。结果与模型对照组比较,异功散组的肺组织损伤减轻;与空白对照组比较,模型对照组和异功散组BALF中IL-10、TGF-β1均显著降低(P<0.05),而IL-17则显著增高(P<0.05);与模型对照组比较,异功散组IL-10显著增高(P<0.05),IL-17降低(P<0.05),而TGF-β1则无显著差异(P>0.05)。结论异功散可减轻哮喘模型幼鼠的气道炎症,提高肺泡灌洗液中IL-10、减少IL-17的含量。
Objective To study the effects of early intervention with Yi Gong powder on airway in-flammation in bronchial asthma rats, in order to explore the mechanism of traditional Chinese medicine in preventing and treating bronchial asthma. Methods 24 female BALB/ c mice of 7 days old were selected, and the mice were randomly divided into the blank control group, the model control group and the Yi Gong powder group. Asthma model of young mice was induced by ovalbumin. The general situation, pulmonary pathology(HE stain), and the changes of interleukin 10(IL-10), interleukin 17(IL-17) and transforming growth factor-beta1(TGF-β1) in bronchial alveolar lavage fluid( BALF) was observed. Results (1) Compared with model group, the lung tissue injury was alleviated in the Yi Gong powder group(P〈0. 05). (2)Compared with the blank control group, IL-10 and TGF-β1 was markedly decreased in the model control group and the Yi Gong powder group ( P 〈 0. 05 ) while the IL-17 were markedly increased (P〈0. 05). Compared with model control group, IL-10 of the Yi Gong powder group was markedly increased(P〈0. 05) while the IL-17 was markedly decreased(P〈0. 05), and there was no significance in TGF-β(P〉0. 05). Conclusion Yi Gong powder can increase the content of IL-10, and decrease the content of IL-17 in bronchoalveolar lavage fluid of asthmatic mice.
