摘要
以黑枸杞为材料,运用pH示差法测定黑枸杞中的花青素。通过对测定波长、缓冲液pH(环境pH)、平衡温度和平衡时间影响因素的研究选择pH示差法最优实验参数:测定波长为525nm,吸光度测定时的缓冲液pH为1.0和4.5,平衡温度为40℃。pH=1.0时的平衡时间为30min,pH=4.5时的平衡时间为20min。运用pH示差法测得的黑枸杞花青素含量为60.59mg/L,平均加标回收率为100.32%,说明准确度高,pH示差法方便快捷,成本低。
Anthocyanin in black wolfberry was detected by pH differential method. The wave length, buffer pH, equilibrium temperature and equilibrium time were selected as influence factors in optimizing the experiment. The best conditions were : wave length 525nm, buffer solution pill.0 and pH4.5, temperature 40℃ . The equilibrium time was 30 rain at pH 1.0 and 20 minutes at pH of 4.5. The anthocyanin in black and lycium barbarum was 60.59mg/L and the average recovery rate was 100.32%. The result showed the high accuracy and low cost. It is suitable in the determination of anthocyanin detection in black wolfberry.
