摘要
目的:观察氯吡格雷对感染幽门螺杆菌(Hp)的小鼠胃黏膜损伤的影响。方法:BALB/c小鼠30只,随机分为对照组、Hp组、(氯吡格雷+Hp)组,10只/组。先将Hp组、(氯吡格雷+Hp)组小鼠进行Hp造模,造模完成后,将(氯吡格雷+Hp)组进行氯吡格雷灌胃,Hp组进行生理盐水灌胃,处死小鼠,按Guth法进行大体评分;采用免疫组化法对Claudin、肿瘤坏死因子(TNF-α)进行定性分析。结果:Hp组、(氯吡格雷+Hp)组小鼠胃黏膜大体评分高于对照组(P<0.05),(氯吡格雷+Hp)组大体评分高于Hp组(P<0.05);免疫组化结果表明,与对照组相比,Hp组、(氯吡格雷+Hp)组Claudin低表达,(氯吡格雷+Hp)组表达水平最低(P<0.05);与对照组相比,Hp组、(氯吡格雷+Hp)组TNF-α高表达,(氯吡格雷+Hp)组表达水平最高(P<0.05)。结论:Hp对小鼠胃黏膜有损伤;氯吡格雷加剧Hp感染小鼠胃黏膜损伤;Claudin参与氯吡格雷作用下的Hp感染小鼠胃黏膜损伤的形成,氯吡格雷通过炎症反应因子TNF-α高表达而加重对已感染Hp小鼠的胃黏膜损伤,表明炎症反应在其中起了重要作用。
Objective:To observe the effect of clopidogrel on gastric mueoeal injury in mice infected with Helicohacter pylori (Hp). Methods :30 BALB/c mice were randomly divided into control group, Hp group, clopidogrel + Hp group, 10 mice in each group. Two experimental groups were first made up model of Hp, after the completion of the model, the clopidogrel + Hp group was given intragastric administration of clopidogrel, while the Hp group was given intragastrie administration of saline. Then the mice were killed, Gross score evaluated by Guth method; immunohistechamistry of Clandin and TNF-α for qualitative analysis was conducted. Results : ( 1 ) The gastric mucasa score in mice in the experimental group was higher than that of the control group ( P 〈0.05 ) ; the gross score of the Clopidogrel + Hp group was higher than that of Hp group ( P 〈 0.05 ) ; ( 2 ) Imraunohistochemistry (Clandin) : compared with the control group, Claudin protein in the experimental groups was expressed at low levels ( P 〈 0.05 ) ; the expression of Clopidagrel + Hp group was the lowest ( P 〈 0.05 ) ; (3) lrnmunohistochemistry ( TNF - α ) : compared with the control group, TNF - α in the experimental grups was expressed at high levels ( P 〈0.05 ) ; the expression of Clopidogrel + Hp group was the highest ( P 〈 0.05 ). Conclusion: ( 1 ) Hp can damage to gastric mucosa in mice; (2) Clopidogrel can aggravate the injury of gastric mueesa in mice with HP infection ; (3) The Clandin protein is involved in the formation of gastric mucosal injury in mice infected with HP by clopidogrel; (4) Clopidagrel is a high expression of the inflammatory factor TNF- α, which aggravates the damage to the gastric mucosa of mice infected with Hp, which indicates that inflammation plays an important role.
