转轮状病毒基因椭圆小球藻生产藻粉和油脂条件的优化

摘要 为了解转轮状病毒基因操作对椭圆小球藻产藻粉和油脂的影响并获取最佳培养条件,利用Western Blot检测验证椭圆小球藻生物反应器的成功建立,之后在不同氮源浓度和pH的培养基中培养野生和转基因椭圆小球藻.结果发现,转基因椭圆小球藻可以成功表达外源基因产生目的蛋白,但外源基因的导入确实降低了椭圆小球藻的藻粉和油脂产量.当培养基的氮源质量浓度为3g/L、初始pH值为5时,转基因椭圆小球藻的藻粉和油脂产量均较高. To investigate the effect of rotavirus gene manipulation on the production of algae and oil of Chlorella ellipsoidea,and get the best culture conditions,Western Blot method was adopted to detect and validate the successful establishment of C.ellipsoidea bioreactor. Then,the wild and transgenic C. ellipsoidea were cultured in different nitrogen concentrations at different pH. The results showed that transgenic C. ellipsoidea could express foreign gene and produce target protein successfully,but the introduction of exogenous gene decreased the production of algae and oil indeed. When the concentration of nitrogen source in the medium was 3 g/L and the initial pH value was 5,the productions of algae powder and oil from transgenic C. ellipsoidea were higher.
