摘要
目的观察葛根素对氧诱导大鼠玻璃体血管内皮生长因子(VEGF)和视网膜血小板内皮细胞黏附分子-1(PECAM-1)表达的影响,探讨其对新生血管的抑制作用。方法初生SD大鼠12只,建立氧诱导初生大鼠新生血管模型,随机分成空白组、葛根素小剂量组及葛根素大剂量组,每组4只。通过荧光素眼底血管造影观察各组新生血管渗漏区域改变,免疫荧光染色观察PECAM-1表达,酶联免疫吸附法(ELISA)测定玻璃体VEGF表达量。结果新生血管渗漏程度、PECAM-1、VEGF表达随葛根素剂量增加而减少;与空白组比较,葛根素小剂量组、大剂量组玻璃体液中VEGF表达均明显降低[(56.58±0.89)pg/mL、(50.72±0.67)pg/mL比(89.11±1.93)pg/mL,P<0.01]。结论葛根素可通过抑制VEGF及PECAM-1表达,抑制氧诱导大鼠视网膜新生血管增生。
Objective To investigate the effect of puerarin on the expression of vascular endothelial growth fac tor(VEGF) and platelet endothelial cell adhesion molecule 1(PECAM-1) in hyperoxia-inducedretinopathy of rats.Methods Rat models of hyperoxia-induced neovascularization were established(n=12). Models were divided into blank, small-dose puerarin, and large-dose puerarin groups(n=4, each). The leakage area was observed using fluorescein fundus angiography,the expression of PECAM-1 and VEGF in vitreum was detected by immunofluorescence staining and ELISA, respectively. Results Puerarin administration reduced neovascularization leakage area, inhibited PECAM-1 and VEGF expressions in a dose-dependent manner. Compared with blank, the expression of VEGF in small-dose group and large-dose group were decreased(56.58±0.89pg/m L and 50.72±0.67pg/m L vs 89.11±1.93pg/m L, P0.01). Conclusion Puerarin can inhibit the hyperoxia-induced retinal neovascularization in rats by inhibiting the expression of VEGF and PECAM-1.
