摘要
目的:研究转化生长因子-β2(TGF-β2)在促进胚胎干细胞(ESCs)分化成心肌细胞中对GATA6表达的影响。方法:用"悬滴"法研究ESCs分化成心肌细胞。将ESCs(CGR8)分为对照组、TGF-β2(2 ng/ml)处理组和TGF-β2(10 ng/ml)处理组。第7天测量拟胚体(EBs)的直径,第14天统计贴壁拟胚体自发节律性跳动的百分数,免疫组织化学和免疫印迹检测GATA6的表达变化。结果:TGF-β2(10 ng/ml)可显著促进拟胚体生长,提高贴壁拟胚体跳动的百分数和促进高贴壁拟胚体中GATA6的表达。结论:TGF-β2促进ESCs分化成心肌细胞,可能与其上调GATA6表达相关。
Objective:To investigate the effects of transforming growth factor-β2 (TGF-β2 )on the expression of GATA6 in promotion of cardiogenesis from embryonic stem cells (ESCs).Methods:The methods of "hanging drop"was used to induce ESCs to differentiate into cardiomyocytes.The CGR8 ESCs were divided into 3 experimental groups:control group,TGF-β2 (2 ng/ml)treatment group and TGF-β2 (10 ng/ml)treatment group.On day 7 ,the diameter of embryonic bodies (EBs)was measured.On day 14,the percentage of EB spontaneous beating was counted.The expression of GATA6 was detected by immunofluorescence and Western blotting.Results:TGF-β2 (10 ng/ml)could significantly promote the EB growth,improve the percentage of EB spontaneous beating and enhance the GATA6 expression.Conclusion:GATA6 may be involved in the effects of TGF-β2 on the promotion of cardiogenesis from ESCs.
