IL-17在大鼠异位气管移植的慢性闭塞性细支气管炎中的表达和作用研究 被引量:4

摘要 目的研究通过大鼠建立气管异位移植慢性闭塞性细支气管炎模型并测定不同阶段的白细胞介素17(IL-17)表达,分析其作用。方法 2015年6月至2016年6月采用大鼠建模的方法研究,将清洁级大鼠40对,随机分为两组,每组20对。其中A组采用Wistar大鼠制备供体,采用SD大鼠制备受体;B组均采用SD大鼠制备供体和受体,进行气管移植建模,分别在各时间段采样完成病理组织分析比较,以评估IL-17的表达和作用。结果实验组在第7、14、18天后病理切片上皮发生不同程度丢失,对照组未见明显改变;软骨破坏程度比较:第7天组间比较无显著差异,第14、28天实验组明显高于对照组(P<0.05);淋巴细胞计数比较:实验组在移植后第7、14、28天明显高于对照组(P<0.05);IL-17水平比较:两组在第7、14天组间无显著差异,第28天实验组IL-17水平较对照组明显升高,差异有显著性(P<0.05);且第28天实验组闭塞性细支气管炎发生率(100.0%)高于对照组的(0.00%)。结论通过建立大鼠异位气管移植慢性排斥反应模型,分析监测病理改变,证实异位气管移植后,IL-17的表达与闭塞性细支气管纤维化发展呈现正性相关。 Objective To establish a model of chronic occlusive bronchitis and to determine the expression of IL-17 in different stages through the establishment of trachea heterotopic transplantation in rats. Methods From June 2015 to June 2016,rat modeling was made,40 rats were randomly divided into two groups,each group of 20 pairs. Wistar rats were used as as donor in group A,SD rats as receptor; In group B,the donor and receptor were prepared by SD rats,and the endotracheal transplantation model was performed to evaluate the expression and function of IL-17 by sampling the pathological tissue analysis in each time period. Results The pathological sections of the experimental group were lost in the pathological sections after the 7 d,14 d and 18 d,and the control group showed no significant changes. Cartilage damage comparison: there was no significant difference after 7 d,but the study group in 14 d,28 d was significantly higher than the control group( P〈0. 05). Lymphocyte counts: the experimental group after transplant 7 d,14 d,28 d was significantly higher than the control group( P〈0. 05); IL-17 level comparison: there was no significant difference between the two groups in the 7 d and 14 d,and the IL-17 level was significantly higher than the control group in the 28 d,with significant difference( P〈0. 05). The incidence of occlusive bronchitis( 100. 0%) was higher than that in the control group( 0. 00%). Conclusion Through the establishment of ectopic tracheal transplantation in rats model of chronic rejection,the pathological change were analyzed and monitored,,the expression of IL-17 and occlusive bronchioles fibrosis development is positive correlated after ectopic tracheal transplantation.
