摘要
目的探讨精神分裂症circRNA表达水平及其与患者阴性、阳性症状的关系。方法分别选取5例精神分裂症患者和正常人外周血进行基因芯片筛查,确定9种存在差异表达的circRNA,并在102例精神分裂症患者和103例正常人中进行PCR验证,进一步行精神分裂症阳性和阴性症状量表评估。结果精神分裂症患者外周血circRNA_102101、circRNA_102315、circRNA_104597、circRNA_101835、circRNA_101836的表达水平明显下调(P<0.05或P<0.01)。circRNA_103102、circRNA_103704的表达水平明显上调(P<0.01)。circRNA_102101、circRNA_103102的ΔCt值与阳性症状呈明显正相关(P<0.01或P<0.05)。circRNA_103704与阳性症状、一般精神病理呈明显正相关(P<0.01或P<0.05)。思维障碍与circRNA_102101、circRNA_103102、circRNA_102315、circRNA_103704、circRNA_102802的ΔCt值呈明显正相关(P<0.01或P<0.05)。激活性与circRNA_102101、circRNA_103102、circRNA_104597、circRNA_103704、circRNA_102802的ΔCt值呈明显正相关(P<0.01或P<0.05)。偏执与circRNA_102101、circRNA_103102、ci rcRNA_103704、ci rcRNA_102802的ΔCt值呈明显正相关(P<0.01或P<0.05)。攻击性与ci rcRNA_102101、circRNA_103102、circRNA_103704、circRNA_102802的ΔCt值呈明显正相关(P<0.01或P<0.05)。circRNA_103704进入阳性症状的回归方程(P<0.01);circRNA_103704、circRNA_102315进入一般病理表现的回归方程(P<0.01或P<0.05)。结论circRNA_103704、circRNA_103102与精神分裂症阴性、阳性症状有明显关联,可能是精神分裂症病理过程主导的调控因子。
Objective To explore the correlation of circ RNAs' expression level to the negative-and positive symptoms of patients with schizophrenia(SZ). Methods Gene chip screening was performed with the peripheral blood samples from each five of SZ patients and normal controls. Nine circ RNA s show ing differentiate expression were confirmed, and further verification was done by real-time fluorescence quantitative PCR in 102 SZ patients and 103 normal controls. All the SZ patients were assessed w ith Positive and Negative Symptom Scale(PANSS). Results It was revealed that the ex pression levels of circ RNA_102101, circ RNA_102315, circ RNA_104597, circ RNA_101835 and circ RNA_101836 were significantly down-regulated(P〈0.01 or P〈0.05), and circ RNA_103102 and circ RNA_103704 were up-regulated in SZ group(P〈0.01). The ΔCT value of circ RNA_102101 and circ RNA_103102 was positively correlated to the positive symptoms(P〈0.01 or P〈0.05), and the ΔCT value of circ RNA_103704 also showed positive correlation with positive symptoms and general psychopathological symptoms(P〈0.01 or P〈0.05). The ΔCT values of circ RNA_102101, circ RNA_103102, circ RNA_102315, circ RNA_103704 and circ RNA_102802 were correlated w ith thinking disorder(P〈0.01 or P〈0.05), and the ΔCT values of circ RNA_102101, circ RNA_103102, circ RNA_104597, circ RNA_103704 and circ RNA_102802 were correlated with the activation(P〈0.01 or P〈0.05). The ΔCT values of circ RNA_102101, circ RNA_103102, circ RNA_103704 and circ RNA_102802 were positively correlated with paranoid(P〈0.01 or P〈0.05), and of circ RNA_102101, circ RNA_103102, circ RNA_103704 and circ RNA_102802 were markedly correlated with assault(P〈0.01 or P〈0.05). Therefore, circ RNA_103704 was chosen into regressive equation of positive symptoms(P〈0.01), and circ RNA_103704 and circ RNA_102315 were chosen into regressive equation of general pathological findings(P〈0.01 or P〈0.05). Conclusion The expression levels of circ RNA_103704 and circ RNA_103102 are obviously up-regulated in SZ patients than in normal controls, and markedly correlated with the negative and positive SZ symptoms, so might be the dominant regulatory factors in the pathological process of schizophrenia.
