摘要
目的评价连翘苷对主动脉斑块形成的干预作用。方法采用Apo E-/-小鼠喂养西方膳食饲料的方法建立动脉粥样硬化模型,建模同时用连翘苷低、中、高三个剂量(5 mg·kg-1、15 mg·kg-1、45 mg·kg-1BW)进行灌胃干预,连续12周。12周后,比较各组主动脉斑块面积以及Ac LDL刺激下的骨髓来源巨噬细胞PPAR-r、ABCA1、ABCG1、SR-B1基因的表达情况。结果与模型对照组相比,连翘苷中、高剂量组的主动脉斑块面积减少(P<0.05),骨髓来源巨噬细胞PPAR-r、ABCA1、ABCG1、SR-B1基因表达均上调(P<0.01,P<0.05)。结论连翘苷具有一定的抗动脉粥样硬化作用,其机制可能与促进巨噬细胞胆固醇流出的途径有关。
Objective To evaluate the intervention of Phillyrin on atherosclerotic plaque formation in murine aortas.Methods The mice were fed with Phillyrin( 5 mg·kg-1,15 mg·kg-1,45 mg·kg-1BW) and western-type diet for 12 weeks.Then,aortas were dissected,Oil Red O stained. Bone marrow cells were cultured into macrophages. Macrophages from all groups were treated with 50 μg·ml-1of acetylated low density lipoprotein( acetylated-LDL) at 48 h for PPAR-γ,ABCA1,ABCG1,and SR-B1 mRNA analysis. Results The atherosclerotic lesion areas in the whole aorta of intervention mice had decreased compared with control mice( P 0. 05). The relative quantitative expression of PPAR-γ,ABCA1,ABCG1,and SR-B1 in macrophages had increased compared with control mice( P〈0. 01,P 0. 05). Conclusion Phillyrin may inhibit atherosclerotic plaque formation through increase cholesterol efflux pathways.
