摘要
该试验对福林-酚比色法测定葡萄烈酒中总多酚含量的最佳条件进行了研究。结果表明,最佳检测条件确定为反应温度40℃、显色时间20 min、检测波长736 nm。在此最佳条件下,没食子酸质量浓度在50~400μg/m L范围内与吸光度值线性关系良好(相关系数R2=0.997 4),方法重复性试验和精密度试验结果相对标准偏差(RSD)分别为1.97%和1.17%,回收率为98.61%~103.22%,RSD值为2.34%,表明该方法重复性好、精密度和准确度较高,可用于葡萄烈酒中总多酚含量的检测。
The optimum determination conditions of total polyphenols content in grape spirits by Folin-ciocalteaut method were researched. The results showed that the optimum determination conditions were be identified as reaction temperature 40 ℃, developing time 20 min and detection wavelength 736 nm. Under the optimum conditions, the gallic acid in the range of 50-400 μg/ml had a good linear relationship with the absorbance value (R2=0.997 4). The relative standard deviation (RSD) of method repeatability test and the precision tests results were 1.97% and 1.17%, respectively. The recovery rate was 98.61%-103.22%, RSD was 2.34%, which indicated the method had good repeatability, precision and high accuracy, and could be used for the determination of total polyphenols content in grape spirits.
