HPLC同时测定鸭跖草提取物中咖啡酸、槲皮素-3-O-α-L-鼠李糖苷和异荭草素的含量

Simultaneous determination of caffeic acid,quercetin-3-O-α-L-rhamnoside and isoorientin in extract of Commelina communis L. by HPLC

采用高效液相色谱法同时测定鸭跖草提取物中咖啡酸、槲皮素-3-O-α-L-鼠李糖苷和异荭草素的含量。色谱条件为:色谱柱为Phenomenex C18(250 mm×4.6 mm,5μm)柱,以甲醇(A)-体积分数0.1%甲酸水(B)梯度洗脱:0~17 min,22%A^42%A;17~28 min,42%A^56%A;28~40 min,56%A^90%A,检测波长为350 nm,流速为1 m L/min,柱温为35℃。咖啡酸、槲皮素-3-O-α-L-鼠李糖苷和异荭草素分别在2.16~86.4 mg/L、6.445~257.8 mg/L和3.293~131.7 mg/L范围内线性关系良好,平均回收率分别为98.36%,99.42%和98.00%。该方法准确、可靠、重复性好,可用于鸭跖草提取物的质量评价与控制。

The study uses HPLC method for simultaneous determination of the content of caffeic acid, quercetin-3-O-α-L-rhamnoside and isoorientin in extract of Commelina communis L.. The HPLC analysis has been carried out on Phenomenex CIs column （4.6 mm×250 mm, 5 μm） with methanol（A）-0.1% formic acid（B） gradient elution：0 min - 17 min, 22% A-42% A;17 min-28 min, 42% A ~56% A;28 min-40 min, 56% A -90% A at a flow rate of 1 mL/min. The detection wavelength is 350 nm and the column temperature is 35℃. Good linear relationship has been found in the the range of 2.16 - 86.4 mg/L for caffeic acid, 6.445 -257.8 mg/L for quercetin-3-O-a-L-rhamnoside, 3. 293 -131.7 mg/L for isoorientin. Their average recoveries are 98.36%, 99.42% and 98.00% , respectively. The method is accurate, reliable, and can be used for the quality control and evaluation of extract of Commelina communis L..
