β转化生长因子增强rhBMP-2诱导成骨的剂量依赖性

Dose-dependent Stimulation of Bone Induction by rhBMP-2 Enhanced by TGF-β

目的 :对 β转化生长因子 (TGF β)增强人重组骨形态发生蛋白 2 (rhBMP 2 )诱导成骨的剂量依赖性进行研究。方法 :2 45只BALB/c小鼠随机分 7组 ,每组 3 5只 ,实验组rhBMP 2的剂量均为 0 .5mg ,TGF β的剂量分别为 2、10、2 0、80、2 0 0、40 0 μg。设立单纯rhBMP 2 (0 .5mg)组为对照。各组于术后 3、5、7、9、14、2 1d共 6个时点取材 ,行组织学、X线分析以及钙含量测定 ,观察 7组的诱导成骨情况。结果 :(1)TGF β剂量为 10～ 80 μg时 ,明显增强rhBMP 2的诱导成骨作用。 (2 )TGF β剂量为 2 2 0 0ug时 ,对rhBMP 2的诱导成骨作用无明显影响。 (3 )TGF β剂量为 40 0 μg时 ,明显抑制了rhBMP 2的诱导成骨作用。结论 :TGF β增强rhBMP

Objective:To study the dose dependent stimulation of bone induction by rhBMP 2 enhanced by TGF β.Methods:BALB/c mice (n=245) were divided into seven groups at random,35 in each group.Mice receiving respectively rhBMP 2(0.5mg)and TGF β(2,10,20,80,200,400 ug)served as the subjects and those receiving rhBMP 2(0.5mg)only served as the control.In order to investigate osteoinduction and osteogenesis,histological analysis were performed 3,5,7,9,14 and 21 days after the implantation.Radiographs of new bone in thigh pouches were taken and calcium content was measured 21 days after the implantation.Results:(1)TGF β with doses from 10 to 80 μg enhanced obviously the osteoinduction and osteogenesis by rhBMP 2.(2)No effect is seen with 2 or 200 ug of TGF β.(3)TGF β with dose of 400 ug had a powerful inhibitory effect.Conclusion:The dose dependent effect is seen in the bone induction by rhBMP 2 enhanced by TGF β.
