三七总皂苷对脑出血大鼠神经细胞凋亡及相关基因的影响 被引量:10

摘要 目的:研究三七总皂苷对脑出血大鼠神经细胞凋亡及相关基因的影响.方法:将大鼠随机分为假手术组、模型组和三七总皂苷组,利用胶原酶注射尾状核法制作脑出血模型,利用免疫组化染色法及细胞凋亡染色法检测各组大鼠Bcl-2、Bax、Caspase-3及TUNEL阳性细胞数目.结果:在6 h、2 d、3 d、5 d、7 d共5个时间点,模型组、三七总皂苷组大鼠神经细胞凋亡情况、Bcl-2、Bax、Caspase-3与假手术组相比差异均显著,具有统计学意义(P<0.05);三七总皂苷组大鼠神经细胞凋亡情况、Bcl-2、Bax、Caspase-3与模型组相比差异均显著,具有统计学意义(P<0.05).结论:三七总皂苷治疗可以显著下调Bax、Caspase-3的表达水平,提高Bcl-2的表达水平,减少神经细胞凋亡,降低脑出血后继发性损伤. Aim: To study the effect of panax notoginseng saponins on neuronal apoptosis and related genes in rats with intracerebral hemorrhage. Methods: Rats were randomly divided into sham operation group,model group and panax notoginseng saponins group. The cerebral hemorrhage model was made by carotenoid injection using collagenase injection. Immunohistochemical staining and apoptosis staining were used to detect the Bcl-2,Bax,Caspase-3 and TUNEL positive cells. Results: At 5 h,2 d,3 d,5 d and 7 d,the neuronal apoptosis,Bcl-2,Bax and Caspase-3 in the model group and Panax notoginseng saponins group were significantly different from those in the sham operation group( P 0. 05). The neuronal apoptosis,Bcl-2,Bax and Caspase-3 in the panax notoginseng saponins group were significantly different from those in the model group( P 0. 05),and the difference was statistically significant( P 0. 05). Conclusion: Panax notoginseng saponins can significantly down-regulate the expression of Baxand Caspase-3,increase the expression of Bcl-2,decrease the apoptosis of nerve cells and reduce the secondary injury after intracerebral hemorrhage.
