乳腺癌患者外周血中树突状细胞表面标记CD1a、CD83的研究

摘要 目的:对乳腺癌患者外周血中培养的树突状细胞(dendriticcells,DC)表面分子CD1a、CD83的表达进行研究。方法:采用密度梯度离心的方法分离人外周血中的单个核细胞,用粒细胞-单核细胞集落刺激因子(GM-CSF)、白细胞介素-4(IL-4)、肿瘤坏死因子α(TNF-α)细胞因子组合,刺激DC增殖、分化。用标有荧光素的单抗标记培养细胞,在流式细胞仪上分析所培养的细胞。结果:该细胞表达CD1a、CD83分子,但CD1a、CD83在CD3+T、CD19+B淋巴细胞上也表达。CD1a、CD83在活化的淋巴细胞上表达水平比未活化的高,CD19+B细胞上表达水平比CD3+T细胞高。结论:CD1a、CD83等分子并非DC特有的标记,目前鉴定DC仍然要靠细胞形态、细胞表达CD1a、CD83以及共刺激分子等综合因素来确定DC。 Objective:To research the expression of molecules CD1a、CD83 on the dendritic cells cultured from peripheral blood of patients with breast cancer.Methods:The mononuclear cells of peripheral blood in 32 patients with breast cancer were isolated by Ficoll Hypaque density gradient centrifugation.Cytokines GM CSF(100 ng/ml), IL 4(20 ng/ml), and/or TNF a(20 ng/ml) were used to culture dendritic cells. Cultured cells were analyzed by flow cytometry with labelled monoclonal antibodies.Results:The characteristic phenotypes such as CD1a+,CD40+,CD80+,CD86+ and CD83+ were demonstrated on the surface of cultured cells.All of these molecules are not specific for dendritic cells. CD1a and CD83 molecules can also be expressed on the surface of CD3+ T and CD19+ B lymphocytes.Both CD1a and CD83 were more highly expressed on actived lymphocytes than that of unactived cells.Expression level of these two molecules on CD19+ B cells was higher than that of on CD3+ T lymphocytes.Conclusions:CD1a and CD83 are not specific phenotypes for dendritic cells.And now,the standardization of dendritic cells was still dependent on morphology with combining the expression of CD1a,CD83 and costimulatory molecules.
