摘要
目的探讨CD11b^+/CD66b^-表型髓系细胞与人类结肠癌进展和肝转移的关系。方法采用real-time PCR、Western blotting及免疫组化方法检测正常结肠组织、结肠癌组织及结肠癌肝转移组织中CD11b、趋化因子配体2(CCL2)和趋化因子受体2(CCR2)的转录水平、蛋白翻译水平。免疫共沉淀方法检测CCL2和CCR2间的相互结合关系。结果结肠癌组织、肝转移瘤组织的CD11b mRNA转录水平及蛋白的表达水平较癌旁正常组织高(P<0.05)。肝转移瘤组织中CCL2的mRNA转录水平及蛋白的表达水平明显高于癌旁正常结肠组织及结肠癌组织(P<0.05),而CCR2mRNA转录水平及蛋白表达水平在三者中无统计学差异(P>0.05)。趋化因子配体CCL2和趋化因子受体CCR2之间可相互结合。结论 CD11b表型髓系细胞在结肠癌组织及结肠癌肝转移组织中广泛浸润,并可能在CCL2-CCR2轴的介导下促进结肠癌进展和结肠癌肝转移。
Objective To study the relationship betw een CD11b~+/CD66b^-phenotype myeloid cells and colon cancer progression and liver metastasis. Methods The transcription level,protein translation levels of CD11 b,chemokine C-C motif ligand 2( CCL2) and chemokine C-C motif receptor type 2( CCR2) in colonic carcinoma tissues,colorectal cancer tissues and liver metastasis w ere detected by real-time PCR,Western blotting and immunohistochemistry. The co-immunoprecipitation method w as used to detect the relationship betw een CCL2 and CCR2. Results The expression of CD11 b mRNA in colorectal carcinoma and liver metastasis w as higher than that in normal tissues( P〈0. 05). The level of CCL2 mRNA transcription and protein expression in liver metastasis w as significantly higher than that in normal colonic tissues and colon cancer tissues( P〈0. 05). The level of CCR2 mRNA transcription and protein expression in the three w ere no significant difference( P〈0. 05). Chemokine ligand CCL2 and chemokine receptor CCR2 can be combined w ith each other. Conclusion CD11 b phenotype myeloid cell is extensive infiltration in colon cancer and liver metastasis of colon cancer tissue,and possibly can promote the progression of colon cancer and liver metastasis of colonic carcinoma by mediating w ith CCL2-CCR2 axis.
