摘要
为促进甜菊糖苷提取用甜叶菊新品种选育,以非糖苷类杂质和甜菊糖苷的检出量为指标,比较了各吸附剂用于甜叶菊种质甜菊糖苷高效液相检测用样品前处理的可行性,并建立了有效的高效液相检测用样品前处理方法。结果表明,甜叶菊叶片经50%乙醇超声浸提后,在所调查的几种吸附剂添加处理中,以酸性氧化铝对浸提液中非糖苷类杂质的去除效果最好,且对以RA和STV为代表的甜菊糖苷含量影响小。进一步的研究揭示,酸性氧化铝用于甜叶菊叶片50%乙醇提取液高效液相检测用样品的前处理优化方案为:在甜叶菊叶片的50%乙醇提取液中加入1/5(m:v)酸性氧化铝,40r/min室温振荡30min,可使提取液中非糖苷类杂质检出量减少65.0%以上,且对以RA和STV为代表的甜菊糖苷检出量的影响小于3.0%。
To promote the selection and breeding of new stevia lines for detection quantity of non-glycosides and SGs were employed as indicators to used in the stevia sample pretreatment for SGs detection by high performance results showed that, compared to the other adsorbents those addition to the remove most of the non-glycoside impurities in the leaching solution and, on steviol glycosides (SGs) extraction, compare the feasibility of adsorbents liquid chromatography (HPLC). The extraction solution, acid A1203 can the same time, take rebaudioside A (RA) and stevioside (STV) as representative of SGs, only a very slight impact were caused on the contents of SGs. Further analysis revealed that, the optimum pretreatment scheme for the HPLC detection SGs in the 50% ethanol extract of stevia leaves by using of acid A1203 was as follows: After adding 1/5 (m: v) acid A1203 to the 50% ethanol extract of stevia leaves, oscillation at 40 r/min under room temperature for 30 min. Pretreatment by above method, the detection quantity of non-glycoside impurities can be reduced by more than 65.0%, and those of SGs, take rebaudioside A and stevioside as representative, were impacted less than 3.0%.
