异甘草酸镁对肝组织中波形蛋白、Ⅰ型胶原蛋白、α-平滑肌肌动蛋白表达的影响

摘要 目的研究异甘草酸镁抑制肝纤维化进程中肝细胞上皮间质转化作用的机制。方法将60只小鼠随机分为6组,每组10只,分别为空白对照组,模型组,异甘草酸镁低剂量组(4 mg/kg)、中剂量组(16 mg/kg)和高剂量组(32mg/kg),水飞蓟宾组(16mg/kg),其中模型组为运用CCl4。HE染色观察各组肝组织的病理学改变,免疫组化法检测肝组织中波形蛋白(Vimentin)、Ⅰ型胶原蛋白(CollagenⅠ)、α-平滑肌肌动蛋白(α-SMA)的表达,Western Blot检测肝组织中Vimentin、CollagenⅠ和α-SMA的蛋白表达情况。结果病理学结果显示,与空白对照组相比,模型组小鼠肝组织的纤维化程度明显加重,肝脏炎症坏死区及纤维间隔明显增多;异甘草酸镁各剂量组小鼠肝组织纤维化水平均明显低于模型组。免疫组化检测和Western Blot结果均显示,模型组肝细胞Vimentin、CollagenⅠ和α-SMA的表达量明显增强;异甘草酸镁能抑制小鼠肝组织中Vimentin、CollagenⅠ和α-SMA的蛋白表达,且具有一定剂量依赖关系。结论异甘草酸镁对CCl4所致小鼠肝纤维化形成有明显的抑制作用。 Objective To study the mechanism of magnesium isoglycyrrhizinate in inhibiting the interstitial transforma- tion of liver epithelial cells in the process of liver fibrosis. Methods Sixty mice were randomly divided into six groups with ten mice in each group, including namely control group, model group, magnesium isoglycyrrhizinate in low dose group (4 mg/kg), magnesium isoglycyrrhizinate in middle close group (16 mg/kg), magnesium isoglycyrrhizinate in high dose group (32 mg/kg) and silibinin group (16 mg/kg). Mices in the model group used CC14. Pathological changes of liver tissue in all groups were ob- served by HE staining. Vimentin, Collagen I and alpha-SMA in liver tissue were detected by immunohistochemistry. The ex- pression of Western Vimentin, Collagen I and alpha-SMA protein in liver tissue were detected by Western Blot analysis. Resuits The pathological results showed that compared with the control group, the degree of liver fibrosis, inflammation of the liver necrosis and fibrous septa in mices of model group increased significantly; the level of liver fibrosis of mices with different doses of magnesium isoglycyrrhizinate were significantly lower than that of the model group. Immunohistochemical detection and Western Blot analysis showed that the expression of Vimentin, Collagen I and alpha-SMA in liver cells of model group increased obviously; magnesium isoglycyrrhizinate could inhibit the expression of Vimentin, Collagen I and alpha-SMA in liver tissue of mices, and had a certain dose-dependent relationship. Conclusion Magnesium isoglycyrrhizinate has obvious inhibitory effect on hepatic fibrosis induced by CC14 in mice.
