摘要
目的:探讨夏枯草配伍钩藤对自发性高血压大鼠(SHR)血压及心脏的影响。方法:14周龄WKY与SHR大鼠经血压测量后,将10只WKY大鼠作为正常组,50只SHR大鼠依据收缩压差异随机分为模型组、硝苯地平组、夏枯草配钩藤组、夏枯草组、钩藤组,每组10只。正常组和模型组给予等体积的去离子水,硝苯地平组给予硝苯地平缓释片溶液,各中药干预组给予相应剂量的中药浓煎液,连续给药4周。使用Softron BP-98A型大鼠无创尾动脉测压仪测量并记录给药前及给药1、2、3、4周后大鼠尾动脉的收缩压,酶联免疫吸附法(ELISA)测定大鼠血清中ANGⅡ、ET、CGRP的含量,测左心室重量指数,苏木精-伊红染色(HE)观察大鼠心脏形态变化。结果:夏枯草配钩藤组、夏枯草组、钩藤组及硝苯地平组SHR收缩压均低于模型组(P<0.05);硝苯地平组、夏枯草配钩藤组降压效果明显,且夏枯草配钩藤组与硝苯地平组比较,差异无统计学意义(P>0.05);夏枯草配钩藤组的降压效果明显优于夏枯草组与钩藤组。中药干预4周后,夏枯草配钩藤组大鼠血清中ANGⅡ、ET含量及左心室质量指数均低于模型组(P<0.05),CGRP含量高于模型组(P<0.05)。结论:夏枯草配伍钩藤可降低自发性高血压大鼠收缩压,控制由高血压造成的心肌肥厚,其作用机制初步认为与降低血清中ANGⅡ、ET的含量,增加血清中CGRP的含量有关。
Objective: To explore the effects of Xiakucao (Selfheal) combined with Gouteng (Uncaria) on blood pressure and heart in spontaneous hypertension rats (SHR). Methods: According to the difference of systolic pressure for WKY and SHR rats of fourteen weeks old, ten Wistar-Kyoto (WKY) rats were as normal group, and 50 SHR were randomly divided into model group, nifedipine group, selfheal combined with uncaria group group , selfheal group and uncaria group, lO rats in each. The rats were given nifedipine by continuous- ly gavages in nifedipine group; selfheal combined with uncaria group, selfheal group and uncaria group were given selfheal and uncaria oral liquid by continuously gavages. The normal group and Model group were given equal volume of deionized water. The systolic blood pressure of the rat tail artery was measured and recorded before and after administration of 1, 2, 3 and 4 weeks by SoftronBP-98A type non-invasive tail artery pressure measuring instrument. Enzyme-linked immunosorbent assay (ELISA) was used to detect the serum concentration of ANG 11 ,ET and CGRP in four weeks. The left ventricular mass index (LVMI) was measured. Hematoxylin eosin staining (HE) was used to observe the pathological changes of heart tissue. Results: The selfheal com- bined with uncaria group, selfheal group, uncaria group and nifedipine group showed lower systolic blood pres- sure than model group (P〈0.05); The antihypertensive effect of Nifedipine group and selfheal combined with uncaria group was more remarkable. Moreover, there was no statistically significant difference between selfheal combined with unearia group and nifedipine group (P〉0.05). The selfheal combined with uncaria group showed better ntihypertensive effect than selfheal group and uncaria group. After treatment for four weeks, the selfheal combined with uncaria group showed lower content of ANG 11, ET in serum than model group (P〈0.05), while higher content of CGRP than model group (P〈O.05). Conclusion: Xiakucao (Selfheal) combined with Gouteng (Un- caria) could reduce blood pressure and control cardiac hypertrophy caused by high blood pressure in SHR, which might be related to reducing serum ANG 11, ET content and increasing the content of CGRP in serum.
