羚羊角与钩藤联用对高热惊厥致发育期大鼠脑损伤的保护作用 被引量:5

Protective Effects of Lingyangjiao (Antelope Horn) and Gouteng (Uncariae) on Brain Injury Induced by Febrile Seizures during Development in Rats

摘要 目的:探讨羚羊角与钩藤联用对高热惊厥所致发育期大鼠脑损伤的保护作用。方法:筛选50只大鼠随机分为空白组、模型组、羚羊角组、钩藤组、羚羊角-钩藤组,每组10只,各药物组大鼠按10 mL/kg给予相应药液,空白组给予等容积蒸馏水。1次/d,连续21 d。给药60 min后,置于(45±0.5)℃恒温水浴箱热水浴实验,隔日1次,连续10次,惊厥发作后,立即取出,擦干,观察大鼠发生惊厥的潜伏时间及惊厥发作持续时间。末次给药24 h后,各组取5只大鼠麻醉处死,取海马进行HE染色并于光镜下观察。结果:羚羊角-钩藤组、羚羊角组及钩藤组在高热惊厥发生后海马CA1区神经细胞病理损伤程度有所减轻,其中羚羊角-钩藤组减轻更明显,优于羚羊角组及钩藤组;羚羊角组及钩藤组惊厥潜伏时间明显延长,且惊厥持续时间明显缩短,与单药组羚羊角组及钩藤组相比,在延长惊厥潜伏时间及缩短惊厥持续时间方面均有明显的优势。结论:羚羊角与钩藤联用在改善脑组织损害方面具有协同作用,且明显优于单药羚羊角组与钩藤组。 Objective: To study the protective effect of Lingyangjiao (Antelope horn) and Gouteng (Uncariae) on brain injury induced by febrile seizures in rats. Methods: The 50 rats were randomly divided into blank group, model group, antelope horn group, uncaria group, antelope horn and uncaria group, ten rats in each. The rats were given liquid medicine with 10 mL/kg in each group, and the blank group was given the same volume of distilled water, 1 times/day, for 21 days. The rats were placed in (45±0.5)℃ constant temperature water bath hot water bath test, 60 rain after administration, every other day, 10 consecutive times; We wiped and dried the rats immediately after seizures, and the latent time of seizures was observed in rats and seizures duration. 24 h after the last administration, 5 rats in every group were sacrificed under anesthesia, and hippocampal tissue was observed under light microscope and HE staining. Results: Antelope horn-uncaria group, antelope horn group and uncaria group in the high fever seizures occurred after hippocampal CA1 neuronal pathological damage has been reduced, which antelope horn group and uncaria group were significantly longer than those in the antelope horn group and the uncaria group. There was a significant advantage in prolonging seizure latency and shortening seizure duration, compared with monotherapy group antelope horn and uncaria group. Conclusion: Lingyangjiao (Antelope horn) and Gouteng (Uncariae) in the improvement of brain tissue damage has synergistic effect, and significantly better than the single antelope horn group and uncaria group.
